An Enzymatic Biosensor for the Detection of D-2-Hydroxyglutaric Acid in Serum and Urine

Summary: In this study, a D-2-hydroxyglutarate dehydrogenase from Ralstonia solanacearum was cloned and recombinantly expressed for the construction of a portable biosensor for D2HG detection. The biosensor showed high sensitivity, a wide detection range, and good stability, indicating its great potential for the diagnosis and treatment of D-2-hydroxyglutaric aciduria and relevant cancers. Detection in complex samples like fetal bovine serum and artificial urine also showed promising results.

Single-cell profiling of D-2-hydroxyglutarate using surface-immobilized resazurin analogs
