Molecular mechanisms of coronary artery disease risk at the PDGFD locus

Hyun-Jung Kim et al.

Summary: Genes encoding risk for coronary disease are crucial for developing new therapies. A particular gene, PDGFD, has been identified as a culprit gene and it promotes disease risk. Mechanistic studies confirm that rs2019090 and PDGFD represent the functional genetic variant and gene responsible for coronary artery disease. PDGFD mediates CAD risk by promoting deleterious phenotypic changes in smooth muscle cells and an inflammatory response primarily focused in the adventitia.

C-type lectin receptor CLEC4A2 promotes tissue adaptation of macrophages and protects against atherosclerosis

Inhye Park et al.

Summary: CLEC4A2 is a distinguishing feature of vascular resident macrophages that have athero-protective properties. It promotes monocyte-to-macrophage differentiation and is critical for maintaining vascular homeostasis. CLEC4A2 deficiency leads to dysfunctional cholesterol metabolism and worsens the development of atherosclerosis.

Summary: This study reveals that TN-X is a central mediator of flow-induced inhibition of EndMT, endothelial inflammation, and atherogenesis by binding to and blocking the activity of TGF-beta. The research uncovers a novel mechanism of flow-dependent regulation of vascular TGF-beta, which holds promise for generating new strategies to prevent vascular inflammation and atherosclerosis.

Embryologic Origin Influences Smooth Muscle Cell Phenotypic Modulation Signatures in Murine Marfan Syndrome Aortic Aneurysm

Albert J. Pedroza et al.

Summary: This study provides a detailed characterization of aortic root smooth muscle cells (SMCs) in Marfan syndrome (MFS), revealing the involvement of SMCs derived from both the second heart field (SHF) and neural crest in the formation of aortic root aneurysms. The study also highlights the subtly distinct transcriptional responses between these two lineages and suggests that TWIST1 may play a key role in the enriched collagen synthesis in SHF-derived SMCs in MFS.

Dichotomous Roles of Smooth Muscle Cell-Derived MCP1 (Monocyte Chemoattractant Protein 1) in Development of Atherosclerosis

Katherine M. Owsiany et al.

Summary: This study investigates the contribution of MCP1 produced by classical smooth muscle cells (SMCs) and Lgals3-transitioned SMCs in atherosclerosis. The results show that MCP1 produced by classical SMCs has an atheroprotective effect, while MCP1 produced by Lgals3-transitioned SMCs exacerbates plaque pathogenesis. These findings highlight the need for caution when considering therapeutic interventions involving MCP1.

Multiple cell types contribute to the atherosclerotic lesion fibrous cap by PDGFR beta and bioenergetic mechanisms

Summary: The study found that stable atherosclerotic plaques are composed of cells from various origins, including smooth muscle cells, endothelial cells, and macrophages that undergo different transitions to produce myofibroblast-like cells. Additionally, it was shown that myofibroblasts derived from endothelial-to-mesenchymal transition and macrophage-to-mesenchymal transition cannot indefinitely compensate for the loss of smooth muscle cell-derived myofibroblasts. Furthermore, the transitions from smooth muscle cells to myofibroblasts are induced by PDGF and TGF-beta, while endothelial-to-mesenchymal transition is induced by interleukin-1 beta and TGF-beta.
