CD47-Dependent Regulation of Immune Checkpoint Gene Expression and MYCN mRNA Splicing in Murine CD8 and Jurkat T Cells

Summary: Elevated expression of CD47 is associated with poor survival in some cancers, but improved survival in cutaneous melanomas. CD47-deficient T cells showed altered gene expression and alternative splicing, including upregulation of MYCN mRNA. Thrombospondin-1 inhibited the expression of MYCN isoforms, as well as the induction of T cell transcripts, resulting in improved overall survival in melanoma patients.

Lacrimal Gland Epithelial Cells Shape Immune Responses through the Modulation of Inflammasomes and Lipid Metabolism

Summary: Lacrimal gland inflammation in dry eye disease is triggered by impaired tear secretion and aberrant inflammasome activation. This study investigated inflammasomes during acute and chronic inflammation and their potential regulators. The activation of inflammasomes in lacrimal gland epithelial cells was induced by bacterial infection and interleukin-1 alpha injection. Chronic inflammation in Sjogren's syndrome models upregulated inflammasome sensors and interleukins, and increased IL-1 beta maturation was observed. Furthermore, altered lipid metabolism and sustained inflammasome activation were identified as key players in the pathogenesis of Sjogren's syndrome-like symptoms.
