Per- and poly-fluoroalkyl substances (PFAS) and female reproductive outcomes: PFAS elimination, endocrine-mediated effects, and disease

Brittany P. Rickard et al.

Summary: Per- and poly-fluoroalkyl substances (PFAS) are environmental contaminants widely detected in drinking water supplies globally, linked to adverse reproductive health outcomes in women. Research has shown that PFAS can disrupt women's reproductive function, but further studies are needed to fully understand the effects on female reproductive health.

Perfluorooctanoic acid alternatives hexafluoropropylene oxides exert male reproductive toxicity by disrupting blood-testis barrier

Bi-Xia Peng et al.

Summary: In this study, the male reproductive toxicity and underlying mechanisms of hexafluoropropylene oxide (HFPO) homologues on blood-testis barrier were explored. The results showed that HFPO homologues and perfluorooctanoic acid (PFOA) disrupted the blood-testis barrier by upregulating the p-p38/p38 MAPK/MMP-9 pathway and promoting the degradation of occludin, a tight junction protein.

Male reproductive toxicity of perfluorooctanoate (PFOA): Rodent studies

Zhongyuan Wang et al.

Summary: The study found that exposure to PFOA leads to damage in the male rodent reproductive system, characterized by abnormal hormone levels, disrupted development of reproductive organs, and abnormal sperm parameters. However, the mechanisms by which PFOA induces these changes have not been identified and require further research.

Male reproductive toxicity involved in spermatogenesis induced by perfluorooctane sulfonate and perfluorooctanoic acid in Caenorhabditis elegans

Jiechen Yin et al.

Summary: PFOS and PFOA exposure was found to reduce brood size, germ cell number, spermatid size, and motility, as well as increase malformation rate of spermatids. PFOS showed higher levels of damage than PFOA in germ cells at lower exposure levels, potentially disrupting various stages of spermatogenesis. Loss-of-function studies revealed that spe-10 gene may play a specific role in PFOS- or PFOA-induced reproductive toxicity, providing valuable data for risk assessment of these pollutants.

PFAS Exposure Pathways for Humans and Wildlife: A Synthesis of Current Knowledge and Key Gaps in Understanding

Amila O. De Silva et al.

Summary: The understanding of human and wildlife exposures to PFAS has mainly focused on a few traditional substances, but there is a need for assessment of a wider range of new PFAS. Diet is a major pathway of human exposure to some PFAS, but there is significant variability across populations and compounds. Total fluorine data and information on unidentified organofluorine are essential, and water has been identified as a major exposure source in contaminated communities. As water sources are remediated, other sources of exposure such as dust, personal care products, and indoor environments may become more significant.

Decabromodiphenyl ether induces male reproductive toxicity by activating mitochondrial apoptotic pathway through glycolipid metabolism dysbiosis

Yue Zhang et al.

Summary: The study revealed that BDE-209 reduced sperm quality and quantity, which was correlated with glycolipid metabolism dysbiosis of testis. Additionally, BDE-209 exposure activated glycolipid metabolism pathways and the mitochondrial apoptotic pathway, leading to spermatogenic cell apoptosis.

Associations of paternal and maternal per- and polyfluoroalkyl substances exposure with cord serum reproductive hormones, placental steroidogenic enzyme and birth weight

Qian Yao et al.

Summary: Maternal PFAS levels were associated with fetal steroid hormones and placental enzymes, while paternal PFAS levels showed no association with these outcomes. The findings suggest that intrauterine PFAS exposure effect on fetal endocrine hormones and growth is unlikely to be confounded by exposure sources or familial factors shared within the couples.

Oxidation of Sperm DNA and Male Infertility

Leila Rashki Ghaleno et al.

Summary: Oxidative stress is a major cause of male infertility due to its destructive effects on sperm structures and functions. Sperm DNA is highly susceptible to oxidative damage, particularly leading to single- or double-strand DNA breaks, which is a significant issue in male infertility. While methods to detect sperm DNA fragmentation have become important diagnostic tools, there is still no clear consensus on their prognostic value.

Biotransformation of perfluoroalkyl acid precursors from various environmental systems: advances and perspectives

Wenping Zhang et al.

Summary: Perfluoroalkyl acids (PFAAs) are widely used due to their unique physicochemical properties, but they are environmentally persistent, toxic, and bioaccumulative. Exposure to PFAAs may arise from direct emission or via the environmental release and degradation of PFAA precursors. Studies on the biotransformation of PFAA precursors have shown that it involves the cleavage of carbon-fluorine bonds and degradation of functional groups.

Signaling Proteins That Regulate Spermatogenesis Are the Emerging Target of Toxicant-Induced Male Reproductive Dysfunction

Sheng Gao et al.

Summary: Emerging evidence suggests that environmental toxicants like cadmium and PFOS may disrupt Sertoli cell and testis function through signaling proteins and cascades similar to pharmaceuticals. This indicates that toxicants may be interfering with physiological mechanisms that support spermatogenesis. Manipulating the expression of these signaling proteins could potentially manage toxicant-induced male reproductive dysfunction.

Perfluoroalkyl Chemicals and Male Reproductive Health: Do PFOA and PFOS Increase Risk for Male Infertility?

Summary: PFAS are synthetic chemicals that have been in existence for over 70 years and are currently being phased out. Studies indicate that PFAS exposure is associated with endocrine disruption on the reproductive system, exhibiting non-monotonic dose responses.

Silver nanoparticles offer a synergistic effect with fluconazole against fluconazole-resistant Candida albicans by abrogating drug efflux pumps and increasing endogenous ROS

Dongmei Jia et al.

Summary: The study found that silver nanoparticles worked synergistically with fluconazole against fluconazole-resistant Candida albicans, both planktonic cells and biofilms formed in less than 12 hours. Silver nanoparticles treatment down-regulated certain genes associated with fluconazole resistance, decreased membrane ergosterol levels and fluidity, reduced membrane content of certain proteins, and elevated ROS production, all contributing to the synergistic effect. In vivo, the combination of silver nanoparticles and fluconazole significantly reduced fungal burden and improved survival rates in infected mice, indicating a promising strategy for treating fluconazole-resistant fungal infections.
