Insights into methanogenesis of mesophilic-psychrophilic varied anaerobic digestion of municipal sludge with antibiotic stress

Summary: This study investigates the influence of temperature variations and antibiotic stress on methane production in psychrophilic anaerobic digestion systems. The results show that methane production is initially inhibited in the presence of oxytetracycline (OTC) under low temperature conditions, but stimulated at 35°C. Methane production and microbial community acclimation of five manure inocula during psychrophilic anaerobic digestion of swine manure

Summary: For small-scale farms, the development of rustic and cheap psychrophilic anaerobic digestion systems is an opportunity to treat manure, mitigate emissions, and promote renewable energy production. In this study, the most efficient inoculum for the start-up of psychrophilic anaerobic digestion of swine slurry at 13 ℃ was found to be swine manure stored for 2 months. After acclimation, the reactor produced a high methane yield, with a production efficiency of 55-68% at 13 ℃ compared to 37 ℃.

Opportunities and challenges of using metagenomic data to bring uncultured microbes into cultivation

Summary: Although we have gained extensive understanding of the diversity of microbial life on earth through culture-independent metagenomic DNA sequence analyses, the isolation and cultivation of microbes is still crucial for direct study and confirmation of their functions and roles. However, the majority of environmental microbes remain uncultured, making it a priority to culture these rare or poorly characterized groups in order to further understand microbiome functions. Additionally, cultivated isolates may have various applications such as new probiotics, biocontrol agents, and industrial processes. The increasing availability of metagenomic and meta-transcriptomic sequence information allows for more opportunities to guide the isolation and cultivation of microbes of interest.

Co-occurrence network analysis reveals loss of microbial interactions in anaerobic digester subjected to repeated organic load shocks

Summary: Fluctuations in the organic loading rate (OLR) of anaerobic digestion (AD) can lead to unstable methane production. This study investigated the impact of repeated high OLR shocks on the microbial community dynamics within AD. The results showed that less frequent organic load shocks led to a decrease in methane productivity and a breakdown of the microbiome network structure. The identification of influential species in the network structure revealed the importance of Anaerovorax, Spirochaetales, and Synergistales. It was inferred that the OLR shocks shifted the microbiome activity away from methane production pathways. Overall, developing a resilient AD microbiome would be necessary for small-scale AD systems that can withstand infrequent organic loading shocks.
