In this study, an asymmetric Satoh-Miura reaction using a SCpRh-catalyst is reported, and it is found that choosing the counteranion of the catalyst is crucial for the desired reactivity.

Transition-metal-catalyzed C-H activation is a powerful strategy for C-C bond formation, but its uptake in total synthesis has been tepid due to synthetic intractability and a lack of comprehensive guidelines. This Review addresses these issues and offers a guide to identify retrosynthetic opportunities to generate C-C bonds by C-H activation processes. Comparing traditional approaches and recent C-H activation methods, this Review demonstrates more efficient retrosynthetic strategies enabled by C-H activation for C-C bond construction.

The reaction success relies on the use of both N-alkoxypyridinium salts and sulfur ylides as radical precursors, allowing for the modular synthesis of various 2,3-disubstituted dihydrobenzofurans with good functional group tolerance, easily available starting materials, simple operation, and mild reaction conditions. Mechanistic studies have shed light on some aspects associated with the key radical intermediates.

Enantioselective Intermolecular Mannich-Type Interception of Phenolic Oxonium Ylide for the Direct Assembly of Chiral 2,2-Disubstituted Dihydrobenzofurans

Summary: An enantioselective intermolecular Mannich-type interception of phenolic oxonium ylides with imines has been developed using cooperative catalysis with achiral dirhodium complex and chiral phosphoric acid. The synthesis of enantioenriched 2,2-disubstituted dihydrobenzofurans with good to high yield and high to excellent enantioselectivity under mild conditions was achieved. Preliminary antitumor activity study showed high anticancer potency of the reduction product 7 against human lung adenocarcinoma cells (A549 cells, IC50 = 9.13 mu M).

Rh(iii)-Catalyzed and synergistic dual directing group-enabled redox-neutral [3+3] annulation of N-phenoxyacetamides with α-allenols
