Ontogeny and Vulnerabilities of Drug-Tolerant Persisters in HER2+ Breast Cancer

Summary: Resistance to targeted therapies in HER2-positive breast cancer is often attributed to the presence of drug-tolerant persisters (DTPs). This study reveals that HER2 TKI treatment leads to the emergence of DTPs with different transcriptomes, and these DTPs are originated from pre-DTP cells that cycle stochastically. The findings provide insights into the ontogeny of DTPs and potential vulnerabilities for therapeutic targeting.

Epigenetic encoding, heritability and plasticity of glioma transcriptional cell states

Summary: This study reveals the key switches for state transitions in diffuse gliomas, recapitulating neurodevelopmental trajectories and highlighting dysregulated epigenetic mechanisms underlying gliomagenesis. The work also demonstrates heritability of malignant cell states and differences in cell state architectures in different types of glioma. Additionally, the study links tumor cell transcriptional states to epigenetics and shows the heritability and importance of these states for tumor plasticity.

Summary: EpCAM(lo) cells are a subpopulation of colon cancer cells with phenotypic plasticity, exhibiting motility, invasiveness, chemoresistance, and high metastatic potential. These cells show enhanced Wnt/beta-catenin signaling, a spectrum of EMT activation states, and a high correlation with the poor-prognosis CMS4 subtype.
