Initiation of translation on nedicistrovirus and related intergenic region IRESs by their factor-independent binding to the P site of 80S ribosomes

TORC1 phosphorylates and inhibits the ribosome preservation factor Stm1 to activate dormant ribosomes

Summary: Target of rapamycin complex 1 (TORC1) promotes ribosome biogenesis and inhibits degradation in response to nutrient availability. The dormant 80S ribosomes formed under nutrient-limited conditions are regulated by the ribosome preservation factor Stm1, which is inhibited by TORC1 upon nutrient replenishment to reactivate translation. Furthermore, the mammalian ortholog of Stm1, SERBP1, is also required for the formation of dormant ribosomes in mammalian cells upon mTORC1 inhibition, suggesting an evolutionarily conserved mechanism for ribosomal dormancy regulation.

Petabase-scale sequence alignment catalyses viral discovery

Summary: Public databases contain a vast amount of nucleic acid sequences, but efficient methods for systematic exploration of this library have been lacking. In this study, we developed a cloud computing infrastructure, Serratus, to enable ultra-high-throughput sequence alignment at the petabase scale. By searching millions of diverse samples, we identified over 10(5) novel RNA viruses and characterized their environmental reservoirs. To facilitate viral discovery, we established a comprehensive database of these data and tools. Expanding the known sequence diversity of viruses has important implications for understanding emerging pathogens and improving pathogen surveillance.

Horizontal gene transfer as a mechanism for the promiscuous acquisition of distinct classes of IRES by avian caliciviruses

Summary: Unlike Picornaviridae members, which have long 5 ' untranslated regions (5 'UTRs) with internal ribosomal entry sites (IRESs), Caliciviridae members typically have short 5 'UTRs and translation initiation is mediated by the interaction between the viral 5 ' terminal genome-linked protein (VPg) and eIF4F subunits instead of IRES. However, it was unexpected to find calicivirus genomes with unusually long 5 'UTRs, prompting a detailed examination. Sequence analysis and structural modeling revealed that these atypically long 5 ' UTRs of Caliciviridae isolates contain picornavirus-like type 2 IRESs, offering evidence for a common evolutionary origin between calicivirus and picornavirus IRESs.

Picorna-Like Viruses of the Havel River, Germany

Expanding known viral diversity in plants: virome of 161 species alongside an ancient canal

Summary: A study was conducted to investigate the virome in a riverside ecosystem, and 245 distinct plant-associated virus genomes were identified. Some of these virus genomes diverged enough to potentially form new species, genera, families, or orders. The study revealed a wider host range for certain viruses than previously recognized, and also the possibility of plant contaminating organisms being the source of some viruses.

RNA viromes from terrestrial sites across China expand environmental viral diversity

Summary: Environmental RNA viruses are diverse, ubiquitous, and have significant ecological and biogeochemical impacts. Metatranscriptomic sequencing of soil, sediment, and animal faecal samples in China revealed a previously unknown diversity of 6,624 putatively novel virus operational taxonomic units. Factors such as organic content and eukaryote species abundance influenced virome compositions, with a greater impact on plants, fungi, and bacteria viruses compared to animal viromes.
