Efficient synthesis, antimicrobial and molecular modelling studies of 3-sulfenylated oxindole linked 1,2,3-triazole hybrids

Vijay Kumar et al.

Summary: A series of novel 3-sulfenylated oxindole linked 1,2,3-triazole hybrids were efficiently synthesized from 3-sulfenylated oxindoles bearing a terminal alkyne. The antimicrobial properties of the synthesized derivatives were evaluated against bacterial and fungal strains, and compound 4j exhibited remarkable efficacy against several strains. Molecular docking studies showed that compound 4j had the best binding affinity with two receptors.

Electrochemical Umpolung C-H Functionalization of Oxindoles

Miryam Pastor et al.

Summary: The study introduces a general electrochemical method for accessing unsymmetrical 3,3-disubstituted oxindoles by direct C-H functionalization, with the oxindole fragment acting as an electrophile. This Umpolung approach eliminates the need for stoichiometric oxidants and operates under mild, environmentally friendly conditions. Importantly, it enables functionalization of these scaffolds via C-O bonds, and can be extended to C-C or C-N bond formation.

Simultaneous ultrasound- and microwave-assisted one-pot 'click' synthesis of 3-formyl-indole clubbed 1,2,3-triazole derivatives and their biological evaluation

Jaydeep A. Mokariya et al.

Summary: An environmentally friendly and high yielding one-pot protocol has been developed for the click reaction of N-propargyl-3-formylindole, chloroacetic acid/ester, and sodium azide using CuI catalyst under simultaneous ultrasound and microwave irradiation. The protocol aims to eliminate the handling of organic azides, making the click process user-friendly and safe. The synthesized compounds show inhibitory action against various pathogenic strains, with some exhibiting excellent potency against E.Coli and P. Aeruginosa.

Diversified strategy for the synthesis of DNA-encoded oxindole libraries

Xuan Wang et al.

Summary: DNA-encoded library technology (DELT) uses DNA as a barcode to track chemical reaction sequences, enabling the design and synthesis of vast libraries of small molecules. This technology has been successfully applied in hit identification and target validation for various diseases. By developing novel on-DNA transformations based on oxindole scaffolds, diverse DNA-encoded libraries are constructed, which have shown promising results in hit identification for protein targets. The diversified synthesis strategy for DEL synthesis has the potential to accelerate the application of synthetic chemistry in drug discovery.
