We have reached a dead end for preimplantation genetic testing for aneuploidy

Summary: The hypothesis of preimplantation genetic testing for aneuploidy (PGT-A) was proposed 20 years ago, but recent studies have shown that its application does not have significant advantages and may even result in adverse outcomes when patient selection biases are reversed. Additionally, high embryo mosaicism leads to the unnecessary discarding of normal embryos, causing harm to patients. Therefore, it is recommended to restrict PGT-A to research protocols and to encourage professional societies to issue appropriate practice guidelines.

Summary: This study investigated how the attributes of mosaicism identified during preimplantation genetic testing for aneuploidy are related to clinical outcomes, and formulated a ranking system for mosaic embryos. The results showed that euploid embryos had significantly better outcomes compared to mosaic embryos, with outcomes influenced by the level and type of mosaicism. Combining mosaic level, type, and embryo morphology revealed a ranking of subcategories regarding the likelihood of positive outcome.

Extensive phylogenies of human development inferred from somatic mutations

Summary: This study reconstructed phylogenies of normal tissues from three adult individuals using whole-genome sequencing, revealing asymmetries in cell contributions and spatial embryonic patterning within and between individuals. The research also showed a spatial effect in brain development and suggested extraembryonic contributions to primordial germ cells. Despite reaching the same ultimate tissue patterns, early bottlenecks and lineage commitments lead to substantial variation in embryonic patterns.
