Aberrant maturation and connectivity of prefrontal cortex in schizophrenia-contribution of NMDA receptor development and hypofunction

Wen-Jun Gao et al.

Summary: The neurobiology of schizophrenia encompasses genetic basis, neurochemical and neurophysiological changes, as well as systemic neural circuitry. The aberrant maturation and connectivity of the prefrontal cortex are key factors in the complex symptoms seen in different stages of the disease.

Fluoxetine treatment supports predictive validity of the three hit model of depression in male PACAP heterozygous mice and underpins the impact of early life adversity on therapeutic efficacy

Tamas Gaszner et al.

Summary: According to the three hit concept of depression, the interaction of genetic predisposition, altered epigenetic programming, and environmental stress factors contribute to the disease. In this study, fluoxetine treatment was found to reduce anxiety and depression levels in mice, and to cause functional and morphological changes in certain brain regions. These findings provide further support for the validity of the three hit concept and the animal model.

Prefrontal cortex PACAP signaling: organization and role in stress regulation

Susan E. Martelle et al.

Summary: PACAP is highly expressed in putative pyramidal neurons in the infralimbic cortex of rats, while PAC1 is enriched in interneurons. Infusion of PACAP into the IL alters stress responses, increases anxiety-like behavior, and enhances fear memory retention in rodents.

PACAP orchestration of stress-related responses in neural circuits

Melissa N. Boucher et al.

Summary: PACAP is a pleiotropic polypeptide that activates G protein-coupled receptors and is widely distributed in the nervous system. It plays roles in regulating fear memories, anxiety-like behavior, and emotional pain. Outside of the CNS, PACAP may also participate in stress responses by modulating the HPA axis.
