Ratiometric pH-Responsive 19F Magnetic Resonance Imaging Contrast Agents Based on Hydrazone Switches

Summary: Hydrazone-based molecular switches are efficient ratiometric pH-sensitive agents that can be tracked using F-19 NMR/MRI and H-1 NMR. Changes in structure induced between pH 3 and 4 result in the appearance and disappearance of signals in the H-1 and F-19 NMR spectra, enabling ratiometric pH measurements. The most significant changes are observed in the resonances of the CF3 group with a shift of 1.8 ppm in F-19 NMR and a hydrazone proton shift of 2 ppm in H-1 NMR.
