Melatonin use among children, adolescents, and young adults: a Danish nationwide drug utilization study

Mette Bliddal et al.

Summary: This study provides a detailed description of the use of melatonin in Danish children, adolescents, and young adults from 2012 to 2019. The incidence of melatonin use increased during this period, and a significant proportion of users had concurrent psychopathology. Long-term use of melatonin was most common among the youngest age groups.

Inhibiting nighttime melatonin and boosting cortisol increase patrolling monocytes, phagocytosis, and myelination in a murine model of multiple sclerosis

Majid Ghareghani et al.

Summary: The study found that melatonin functions through its receptors in the brain of multiple sclerosis (MS) patients. Excessive intake of melatonin exacerbates inflammation and immune cell migration to lesion areas. Therefore, monitoring the intake of melatonin in patients is necessary during treatment.

Prince Sebastian et al.

Summary: This study aims to determine the effects of sun exposure and ultraviolet radiation (UVR) exposure on the risk of pediatric-onset multiple sclerosis (MS). The results showed that spending more time outdoors during summer and higher summer ambient UVR dose were associated with a reduced risk of developing MS in children.

Prednisolone induces sleep disorders via inhibition of melatonin secretion by the circadian rhythm in zebrafish

Yu Jiang et al.

Summary: This study investigated the effects of prednisolone (PN) on sleep in zebrafish. The results showed that PN decreased melatonin secretion, shortened sleep time, and disrupted the internal circadian rhythm. The mechanism behind these effects involves the repression of certain genes via the glucocorticoid receptor, leading to a decrease in melatonin secretion and subsequent sleep disorders.

Reduced melatonin levels may facilitate glioblastoma initiation in the subventricular zone

Summary: Melatonin rhythm and its receptor control the day/night difference in neural stem cell division, and the decline in circadian melatonin synthesis and release as well as its receptors in aging contribute to the initiation and growth of glioblastoma.

PDK4 Inhibition Ameliorates Melatonin Therapy by Modulating Cerebral Metabolism and Remyelination in an EAE Demyelinating Mouse Model of Multiple Sclerosis

Majid Ghareghani et al.

Summary: Combination therapy of melatonin and DADA significantly reduced the severity of experimental autoimmune encephalomyelitis (EAE), a mouse model of multiple sclerosis. The therapy improved immune response, promoted oligodendrocyte generation, and affected cerebral energy metabolism. This study highlights the importance of combined treatment for enhancing the efficiency of melatonin in remyelination.

Sunlight exposure exerts immunomodulatory effects to reduce multiple sclerosis severity

Patrick Ostkamp et al.

Summary: The study found that high serum vitamin D levels were associated with reduced MS severity, lower relapse risk, and slower disability accumulation, while low latitude was linked to higher vitamin D levels, lower MS severity, fewer lesions, and less disability accumulation. Sun exposure may have beneficial effects on established MS, but could potentially be harmful for photosensitive individuals. Additionally, UV-mediated immune-modulation in MS may involve direct induction of type I interferons.

The latitude gradient for multiple sclerosis prevalence is established in the early life course

Clive E. Sabel et al.

Summary: The strongest epidemiological evidence for environmental impact on multiple sclerosis risk is the latitudinal gradient, indicating a significant influence of latitude on prevalence, incidence, and mortality worldwide. Early life environmental exposures, including in utero and at birth, drive this gradient, with prevention efforts recommended for high-risk individuals, especially females, from the earliest possible time points.

Melatonin inhibits embryonic rat H9c2 cells growth through induction of apoptosis and cell cycle arrest via PI3K-AKT signaling pathway

Anda Zhao et al.

Summary: This study showed that high levels of melatonin could inhibit cell growth through apoptosis and cell cycle arrest via PI3K-AKT signaling pathway in H9c2 embryonic rat cardiac cells. The findings suggest that melatonin may interfere with embryonic heart development by down-regulating genes related to heart development and activating the PI3K-Akt signaling pathway. Further studies using human embryonic cardiac cells are warranted to confirm these results.
