Summary: Plant specialized 1,4-naphthoquinones are a remarkable case of convergent evolution. This study sheds light on the metabolic connections between shikonin biosynthesis and its precursor pathways, identifies shikonin pathway gene candidates and their evolutionary histories, and establishes additional evolutionary links between shikonin and ubiquinone metabolism. Moreover, it demonstrates the effectiveness of global coexpression analysis using limited transcriptomic data for identifying gene connections within a defined metabolic network.

Summary: Geraniol, citronellol, and their esters are high-value acyclic monoterpenes used in various industries. This study provides evidence that their biosynthesis mainly occurs in the cytosol of glandular trichomes via geranyl monophosphate and Nudix hydrolase. Different Pelargonium chemotypes show varied accumulation of precursors and end products, highlighting the role of the cytosol in monoterpene biosynthesis.
