Isolation of single circulating trophoblasts from maternal circulation for noninvasive fetal copy number variant profiling

Anna Doffini et al.

Summary: This study developed a multi-step workflow for isolating cEVTs using a semi-automated process and a proprietary algorithm for genetic confirmation and CNV detection. The method demonstrated high resolution and accuracy, making it feasible for noninvasive prenatal testing of fetal genomic profiling.

Initial Clinical Experience with NIPT for Rare Autosomal Aneuploidies and Large Copy Number Variations

Thomas Harasim et al.

Summary: This study evaluated the clinical outcome of non-invasive prenatal testing (NIPT) results positive for rare autosomal aneuploidies (RAA) and large copy number variations (CNVs). The study found that 50% of pregnancies with an NIPT-positive result for RAA showed signs of placental insufficiency or intra-uterine death, while the other 50% had a normal outcome at birth. Additionally, CNV testing by NIPT allowed for the detection of unbalanced translocations and relevant maternal health conditions.

Validity and Utility of Non-Invasive Prenatal Testing for Copy Number Variations and Microdeletions: A Systematic Review

Luca Zaninovic et al.

Summary: Sufficient data on prenatal cell-free DNA-based screening tests for copy number variations and microdeletions are still lacking. Different methodological approaches show variations in diagnostic accuracy measurements and positive predictive values. Caution should be exercised in using NIPT for CNVs and microdeletions, and any new technologies should undergo strict evaluation before implementation. Testing indications should be in line with international guidelines in the field of prenatal diagnostics.

National data on the early clinical use of non-invasive prenatal testing in public and private healthcare in Denmark 2013-2017

Ida C. B. Lund et al.

Summary: In Denmark, the usage of NIPT differs between public hospitals and private clinics, with NIPT primarily used after combined first trimester screening in public settings, while 25% of NIPTs in private clinics were performed before gestational week 11. Regardless of indication, the national public sensitivity for detecting chromosomal aberrations was high, with a majority of true-positive NIPT results in public settings resulting in live births.

Overview and recent developments in cell-based noninvasive prenatal testing

Liesbeth Vossaert et al.

Summary: Investigators have been interested in the natural phenomenon of fetal and placental cell trafficking into the maternal circulation. The scarcity of these cells makes their detection and isolation technically challenging, but they have the potential for considerable benefit over cell-free DNA-based NIPT. Endocervical trophoblasts are also being investigated as an approach for cell-based NIPT, with different approaches' advantages and drawbacks being highlighted in studies around the world. Guidance for developing a sound cell-based NIPT protocol is also provided.

Screening for Fetal Aneuploidy and Sex Chromosomal Anomalies in a Pregnant Woman With Mosaicism for Turner Syndrome-Applications and Advantages of Cell-Based NIPT

Line Dahl Jeppesen et al.

Summary: The study compared the differences in detecting fetal sex-chromosomes anomalies between cell-free NIPT and cell-based NIPT, suggesting that cell-based NIPT may offer a non-invasive testing option for screening SCAs in women with mosaicism for monosomy-X in blood.

Trends in Non-invasive Prenatal Screening and Invasive Testing in Denmark (2000-2019) and Israel (2011-2019)

Lena Sagi-Dain et al.

Summary: The study evaluating the trends in NIPS and CMA use in Denmark and Israel found relatively stable rates of invasive testing and a slight decline in NIPS in both countries. It was speculated that Danish and Israeli women and/or health providers prefer the high resolution and yield of CMA testing over the non-invasiveness of NIPS.
