Transcription Factors Pmr1 and Pmr2 Cooperatively Regulate Melanin Biosynthesis, Conidia Development and Secondary Metabolism in Pestalotiopsis microspora

Summary: This study identified two transcription factors, Pmr1 and Pmr2, in the filamentous fungus Pestalotiopsis microspora, and revealed their crucial roles in melanin synthesis, asexual development, and secondary metabolism. Pmr1 played a larger regulatory role in secondary metabolism, while both Pmr1 and Pmr2 were involved in the biosynthesis of conidia pigment.

AaHog1 Regulates Infective Structural Differentiation Mediated by Physicochemical Signals from Pear Fruit Cuticular Wax, Stress Response, and Alternaria alternata Pathogenicity

Summary: In this study, the regulatory role of the Hog1 mitogen-activated protein kinase in the differentiation of the infective structure of Alternaria alternata was investigated. It was found that AaHog1 plays a significant role in the formation of appressorium and infection hyphae, and is beneficial for the growth, stress tolerance, virulence, and cell-wall-degrading enzyme activity of A. alternata. These findings provide important insights into the regulatory mechanism of AaHog1 in the pathogenesis of A. alternata.

Molecular evolution and regulation of DHN melanin-related gene clusters are closely related to adaptation of different melanin-producing fungi

Summary: Many genes responsible for melanin biosynthesis in fungi were physically linked together, and most of the melanin-producing fungi had PKS gene clusters regulated by Cmr1. Close rearrangement of PKS gene clusters had evolved in these fungi, with various beneficial functions of melanin helping them adapt to changing environments. The fungi had undergone at least five large-scale differentiations, leading to rapid evolution of their PKS gene clusters and adaptation to diverse environments. Recent gene losses and expansion in the PKS gene clusters were frequent, indicating rapid evolution and adaptation of the fungi to different environments. Strong co-evolution was observed between PKS gene and CMR1 gene, but horizontal gene transfer events may have occurred in the genome-duplicated species Aspergillus and Penicillium.
