Whole blood transcriptional signatures associated with rapid antidepressant response to ketamine in patients with treatment resistant depression

Flurin Cathomas et al.

Summary: Ketamine has rapid and sustained antidepressant effects in treatment-resistant depression (TRD) patients, although the underlying mechanisms are still unclear. This study found that TRD patients had a gene expression pattern indicative of interferon signaling pathway activation at baseline, and responders to ketamine treatment showed differential gene expression compared to non-responders. However, no gene expression changes related to an anti-inflammatory effect were observed.

Metabolomic signatures associated with depression and predictors of antidepressant response in humans: A CAN-BIND-1 report

Summary: Caspani et al. demonstrate sex-specific plasma lipoproteins that can predict response to common antidepressants. They find that lipoproteins of the same class but different in size and density have unique associations with depression and treatment response, emphasizing the importance of evaluating lipoprotein subclasses in depression research.

Summary: The study found that serum levels of IL-33 and sST2 are associated with the development of depression after acute ischemic stroke. IL-33 and hsCRP are positively correlated with depression, while sST2 is negatively correlated with depression.

Sex differences in the genetic regulation of the blood transcriptome response to glucocorticoid receptor activation

Sarah R. Moore et al.

Summary: Substantial sex differences have been reported in the physiological response to stress, including the release of the stress hormone cortisol. Genomic variants in regulating the transcriptional response to cortisol via glucocorticoid receptor activation were explored in females and males. The study revealed that while transcripts responsive to glucocorticoid receptor activation overlapped between males and females, the regulation of differential transcription to this activation by genetic variants was distinct. Additionally, genetic profiles regulated by sex-specific genetic variants were predictive of depression status in a child and adolescent cohort in a sex-concordant manner.

Higher polygenic risk scores for schizophrenia may be suggestive of treatment non-response in major depressive disorder

Giuseppe Fanelli et al.

Summary: This study investigated the association between polygenic risk scores (PRS) and response to antidepressants in patients with major depressive disorder (MDD). Results showed that PRS did not predict treatment response, but SCZ-PRS was nominally associated with non-response. Patients with higher SCZ-PRS were more likely to be non-responders to antidepressants, indicating a potential genetic liability to schizophrenia affecting treatment response in MDD.

Transcriptomics and sequencing analysis of gene expression profiling for major depressive disorder

Pragya Verma et al.

Summary: The study compared mRNA expression profiles in MDD, MDD-S, and control groups to identify genes associated with depression. It revealed upregulation of PRKACB and downregulation of genes in glutamatergic and dopaminergic pathways in MDD patients. Dysregulated genes in cholinergic synapses were also observed in MDD patients. The findings enhance our understanding of depression etiology and potential biomarkers.

Nicole Mariani

Summary: The development of major depressive disorder is likely a combination of different biological processes, with gene expression playing a key role in treatment. However, selecting the best antidepressant for each individual remains a challenge.
