An experimental strategy to probe Gq contribution to signal transduction in living cells

Summary: The study introduced an experimental strategy to discriminate between on-target and off-target effects of inhibitors by developing a Gαq variant resistant to FR/YM inhibition while maintaining catalytic activity. The findings demonstrated that the cellular effects of FR/YM were specifically mediated via Gαq proteins, and the resistance mutant eliminated non-Gq protein inhibition contributions. The combined application of FR or YM with the drug-resistant Gαq variant proved to be a powerful in vitro strategy for uncovering Gq input to complex biological processes with high accuracy and specificity.
