Vaccinium as a comparative system for understanding of complex flavonoid accumulation profiles and regulation in fruit

This update highlights the significant gains in genomic tools and functional analysis, using Vaccinium as an example.

Summary: Members of the Vaccinium genus, such as blueberry and bilberry, have different concentrations and localization of anthocyanins due to the expression of key regulatory genes MYBPA1.1 and MYBA1. Blueberry lacks VcMYBA1 expression, resulting in insufficient VcMYBPA1.1 for anthocyanin regulation, while bilberry expresses VmMYBA1 which up-regulates VmMYBPA1.1 for coordinated regulation of flavonoid biosynthesis genes and anthocyanin accumulation.

Knockout of phytoene desaturase gene using CRISPR/Cas9 in highbush blueberry

The Coordinated Action of MYB Activators and Repressors Controls Proanthocyanidin and Anthocyanin Biosynthesis in Vaccinium

Summary: This study identified activator and repressor transcription factors (TFs) that regulate the production of anthocyanins, pigment compounds that positively affect human health, in blueberry and bilberry fruits. MYBA1 activates the pathway while MYBC2.1 represses it. Establishment and optimization of agrobacterium-mediated transformation in blueberry (Vaccinium species)

Summary: This study investigates the effects of variety specificity, explant types, and culture schemes on callus development, adventitious bud regeneration, and transformation efficiency in 19 Vaccinium varieties. The diploid wild Vaccinium reticulatum 'Red Button' is identified as an ideal variety for genetic transformation, with a transformation efficiency of about 12.82% for the green fluorescent protein (GFP) encoding gene. In Vitro Polyploid Induction of Highbush Blueberry through De Novo Shoot Organogenesis

Summary: Polyploid induction is important for the development of new varieties in horticultural plants. In this study, a protocol for in vitro polyploid induction of highbush blueberry leaf tissues was studied. Different concentrations of colchicine and oryzalin were used, and it was found that colchicine can induce polyploidization. The resulting polyploids showed morphological differences compared to diploids.

MYBA and MYBPA transcription factors co-regulate anthocyanin biosynthesis in blue-coloured berries

Summary: This study comprehensively characterized the regulatory network of R2R3 MYB transcription factors in anthocyanin biosynthesis in blue berries of bilberry. It identified a new type of co-regulation between MYBA, MYBPA1, and MYBPA2 subgroups, and associated MYBPA3 subgroup with proanthocyanidin biosynthesis. The results suggest gene duplication enabled functional diversification among various MYB subgroups for the intricate regulation of anthocyanin profiles in blue-coloured berries.
