State of the Art of Patient-reported Outcomes in Acromegaly or GH Deficiency: A Systematic Review and Meta-analysis

Summary: This study examined the landscape of patient-reported outcome measures (PROM) and differences between PROMs and biochemical outcomes in patients with acromegaly and growth hormone deficiency. The findings suggest that prospective studies in these patient populations employ a variety of PROMs, often with poor reporting quality. Discrepancies between PROMs and biochemical outcomes were also observed. Therefore, there is a need to harmonize the use of PROMs in clinical practice and research through the development of core outcome sets.

Summary: Factors associated with impaired QoL differed in young and middle-aged patients compared to older patients with acromegaly. In older patients, arthropathy and higher BMI were associated with poor QoL, indicating the importance of early diagnosis and appropriate treatment to prevent arthropathy in acromegaly.

The number of symptoms and comorbidities, gender, radiotherapy, and medical treatment with somatostatin analogs were identified as factors influencing QoL in these patients.

Case Report: Temozolomide Treatment of Refractory Prolactinoma Resistant to Dopamine Agonists

Summary: Therapeutic agents for refractory prolactinomas that are resistant to dopamine agonists are challenging, with surgery often achieving only partial tumor removal. Second-line treatment with temozolomide is effective for about half of patients, but complete remission is rare. This case report discusses a patient with cabergoline-resistant prolactinoma who achieved complete remission with temozolomide therapy after undergoing two transsphenoidal surgeries. Genomic sequencing revealed 16 possible gene mutations, including a missense mutation of the PABPC1 gene.
