Evidence that an Unnamed Isometric Virus Associated with Potato Rugose Disease in Peru Is a New Species of Genus Torradovirus

Summary: A new torradovirus species called potato rugose stunting virus (PotRSV) was detected in potatoes from Peru, the United States, and the Netherlands. The virus has high sequence identity to an unknown isometric virus associated with potato rugose stunting disease in Peru. The genome of PotRSV consists of two RNA segments, with RNA1 encoding a replication-related polyprotein and RNA2 encoding a movement protein and capsid proteins.

Phylogenetics and Evolution of Potato Virus V: Another Potyvirus that Originated in the Andes

Summary: Potato virus V (PVV) originated in the Andes region and only a few isolates spread to other regions. PVV evolved rapidly and shares similar ecological characteristics with Potato virus Y (PVY). Through phylogenetic reconstruction and dating calculations, PVV is estimated to have appeared around 29 BCE and diversified after the potato late blight pandemic in 1853 CE.

Recombination and darwinian selection as drivers of genetic diversity and evolution of sweet potato leaf curl viruses in Tanzania

Summary: Using HTS and Sanger sequencing techniques, this study detected and analyzed the genetic diversity, recombination events, and evolutionary selection pressure of SPLCVs in Tanzania. The results showed high genetic diversity of the detected SPLCVs and their close relationship with other similar viruses. In addition, recombination events were found in SPLCVs and different evolutionary selection pressures were observed in their genes.

Maize miR167-ARF3/30-polyamine oxidase 1 module-regulated H2O2 production confers resistance to maize chlorotic mottle virus

Summary: Maize employs a signaling mechanism mediated by microR167-auxin response factor 3 and 30-polyamine oxidase 1 for defense and counterdefense against maize chlorotic mottle virus. The study showed that Zma-miR167 positively modulates the degree of resistance to MCMV by targeting ZmARF3 and ZmARF30, which negatively regulate resistance. However, MCMV uses p31 protein to counteract Zma-miR167-mediated defense. The results reveal the role of Zma-miR167-ZmARF3/30 module in restricting MCMV infection.

Onsite detection of plant viruses using isothermal amplification assays

Summary: Plant diseases caused by viruses have a significant impact on crop production and quality, resulting in substantial losses. The options for managing viruses are limited due to their nature as systemic obligate parasites. In this regard, sensitive, robust, and affordable diagnostic assays are needed to detect the presence of viruses in plant materials and prevent their spread. Traditional methods based on biological and physical properties have limitations in terms of sensitivity and time consumption. On the other hand, assays based on viral proteins and nucleic acids offer more specificity, sensitivity, and rapidity. However, they often require sophisticated equipment and technical skills in laboratory settings. Isothermal-based assays, such as LAMP and RPA, provide a simple and reliable alternative that does not require specialized equipment or skills. These assays can be performed using lateral flow devices, making them suitable for onsite detection or testing in the field. Isothermal amplification assays can also be combined with CRISPR/Cas technology to overcome non-specific amplification and cross-contamination issues. The collateral activity associated with some CRISPR/Cas systems has been successfully utilized for the visual detection of plant viruses.

The study also discusses the possible exacerbating effects of climate change on MLN and the negative genetic association between tolerance to heat/drought and susceptibility to viral infection. The role of microRNAs in host plant response to MLN and heat/drought stress is also reviewed, offering a novel molecular approach for developing resistant varieties.
