Epigenome plasticity in plants

James P. B. Lloyd et al.

Summary: The authors review the intra-individual and inter-individual plant epigenome variation during development and in response to environmental changes, stress included. They also discuss the functions of epigenome plasticity and editing technologies that will drive future research. Advancements in technologies for reading and writing epigenomic patterns are enabling a more detailed characterization and functional interrogation of plant epigenome variation.

How Stress Facilitates Phenotypic Innovation Through Epigenetic Diversity

Thanvi Srikant et al.

Summary: Plant adaptation to global warming will require short-term adaptive efforts due to the lack of responsiveness of long-term evolutionary mechanisms. Epigenetic regulation of plant genomes and population diversity can be harnessed for shaping co-evolving traits in response to new climate conditions.

A new take on organelle-mediated stress sensing in plants

Isaac J. Dopp et al.

Summary: Plants have the ability to adjust their phenotype in response to environmental changes, which relies on their internal sensing and processing capabilities. Recent studies have shown that mitochondria and plastids serve as important environmental sensors in plants, triggering responses to stressful conditions and modulating gene expression, metabolism, and the energy balance between growth and defense. These findings contribute to a comprehensive hypothesis for the regulation of plant phenotypic plasticity.

The TUTase URT1 connects decapping activators and prevents the accumulation of excessively deadenylated mRNAs to avoid siRNA biogenesis

Helene Scheer et al.

Summary: The Arabidopsis TUTase URT1 interacts directly with the translation inhibitor and decay factor DECAPPING5, preventing the accumulation of deadenylated mRNAs, and thereby suppressing siRNA biogenesis.

The PARP Way to Epigenetic Changes

Simone Ummarino et al.

Summary: ADP-ribosylation is a reversible post-translational modification that modulates essential cellular processes through PARylation catalyzed by specialized enzyme poly ADP-ribose polymerases (PARP). PARP1 plays important roles in chromatin remodeling and epigenetic regulation, with therapeutic implications for cancer treatment and beyond.

Structure and dynamics of the chromatin remodeler ALC1 bound to a PARylated nucleosome

Luka Bacic et al.

Summary: Here, we reconstituted an ALC1 and PARylated nucleosome complex modified in vitro, enabling cryo-EM structure determination to visualize several intermediate states of ALC1. Functional biochemical assays with PARylated nucleosomes highlighted the importance of nucleosomal epitopes for productive remodeling, and suggested that ALC1 prefers sliding nucleosomes away from DNA breaks.

Differences in the Manifestation of Cell Pluripotence In Vivo and In Vitro in the Mutant Arabidopsis thaliana with the Phenotype of Cell Memory Disorder

E. V. Kupriyanova et al.

Summary: In vitro-cultured plant cells provide a convenient model for studying the genetic and physiological mechanisms required for cell pluripotency. Mutant plant cells with higher expression levels of pluripotency genes have increased potential for de novo shoot regeneration, but the tae mutant, although able to resume pluripotency gene expression and cell proliferation in late leaf development stages, showed decreased regenerative potential in cultured cells. This mutant provides a unique model for studying the role of epigenetic modifications in regulating cell pluripotency.
