Simple molecular identification and free amino acid contents of Neoporphyra kitoi (Bangiales, Rhodophyta) with potential use as a new species for nori cultivation

Summary: We conducted preliminary experiments to confirm the growth of gametophytic blades in the nori farm using the OHN-1 strain of Neoporphyra kitoi. The blades, both mature and immature, grew in the farm and were identified as N. kitoi through nucleotide sequencing. PCR-RFLP analyses were developed to differentiate N. kitoi from Neopyropia yezoensis, a species cultivated in Japan, for future growth comparisons. In addition, the analysis of four major FAA contents showed that OHN-1 had higher levels, especially of alanine, compared to HG-4, suggesting its potential as a new cultivated species.

Summary: Neoporphyra kitoi is a newly described species of red algae found in Honshu, Japan. It is distinguished from other similar species by both molecular and developmental characteristics, with the unique feature of new blades arising from rhizoidal cells. Molecular phylogenetic analysis places Neoporphyra kitoi in a clade containing sequences of the genus Neoporphyra.

Summary: The bladed Bangiales collected from Shikinejima Island and Hachijojima Island in southern central Japan were identified as Neoporphyra haitanensis based on morphological and molecular analyses, showing significant similarities to the cultivated species in southern China. This study represents the first record of N. haitanensis in natural populations in Japan.
