Atroposelective Access to 1,3-Oxazepine-Containing Bridged Biaryls via Carbene-Catalyzed Desymmetrization of Imines

Summary: In this study, a novel bridged biaryl containing medium-sized rings was selectively synthesized via N-heterocyclic carbene organocatalysis. The reaction began with the addition of the carbene catalyst to the aminophenol-derived aldimine substrate, followed by oxidation and intramolecular desymmetrization to form 1,3-oxazepine-containing bridged biaryls with high yields and excellent enantioselectivities. These products can be readily transformed into chiral phosphite ligands.

Stereodivergent Access to [6.7]-Fused N-Heterocycles Bearing 1,3-Nonadjacent Stereogenic Centers by Pd-Catalyzed [4+2] Annulations

Summary: We have developed a highly efficient stereodivergent [4 + 2] annulation reaction using palladium catalysis to synthesize N-heterocycles with 1,3-nonadjacent stereogenic centers. The diastereoselectivity of the reaction is dependent on the polarity of solvents used. Additionally, good enantioselectivities were achieved by employing the chiral ligand Wingphos.

Functionalized cyclopentanes can be synthesized with good yields and stereocontrol by intermolecular cycloaddition between reactive intermediates.

Synthesis of 7-Arylthiomethyl Dibenzo[b,d]azepines through Imidoylative Heck Cyclization and CPA-Catalyzed Thio-Michael Addition/Enantioselective Protonation

Summary: In this study, a chiral phosphoric acid-catalyzed thio-Michael addition/enantioselective protonation has been developed for the first time. The reaction uses 7-methylene-6-aryl-7H-dibenzo-[b,d]azepines, products of Pd-catalyzed imidoylative Heck cyclization, as Michael acceptors in reactions with a wide range of aryl thiols. Diversified 7-[(arylthio)methyl]-7H-dibenzo[b,d]-azepines bearing a benzylic stereocenter and a thermodynamically regulated biaryl axis were produced with good to excellent enantioselectivity and 14-25:1 diastereoisomeric ratios.

Palladium-Catalyzed Intermolecular Asymmetric Dearomative Annulation of Phenols with Vinyl Cyclopropanes

Summary: In this study, we describe a Pd(0)-catalyzed intermolecular asymmetric dearomative [3 + 2] annulation of phenols with vinyl cyclopropanes through the in situ generation of ortho-quinone methide intermediates. Highly functionalized spiro-[5,6] bicycles with three contiguous stereogenic centers, including an all-carbon quaternary, were obtained with excellent stereoselectivities. Density functional theory (DFT) calculations suggest that the reactions are controlled by thermodynamics.

Enantioselective Construction of Axially Chiral Azepine-Containing P,N-Ligands from L-Alanine

Summary: A family of axially chiral azepine-containing seven-membered cyclic P,N-ligands, named Indole-azepinap, was prepared using L-alanine as an original chirality source. The direct chromatographic separation of two diastereomeric phosphine oxides on silica gel allowed easy accessibility of these ligands for further structural and electronic modifications. These Indole-azepinaps showed promising results in a Pd-catalyzed asymmetric allylic alkylation with high yields and moderate enantioselectivities.

Palladium-Catalyzed Enantioselective 7-exo-Trig Carbopalladation/Carbonylation: Cascade Reactions To Achieve Atropisomeric Dibenzo[b,d]azepin-6-ones

Summary: Enantioselective 7-exo-trig cyclocarbopalladation-initiated carbonylation cascade reactions have been successfully achieved, yielding seven-membered dibenzo[b,d]azepin-6-ones with a thermodynamically controlled stereogenic axis and excellent diastereo- and enantioselectivities. The calculated energy difference between diastereoisomers is approximately 2.8 kcal/mol, explaining the observed high diastereoselectivity.

Structurally Diverse Synthesis of Five-, Six-, and Seven-Membered Benzosultams through Electrochemical Cyclization

Summary: A metal- and oxidant-free approach has been developed for the synthesis of structurally diverse benzosultams from aryl sulfonamides through an electrochemical cyclization. By varying the ortho substituent on aryl sulfonamides, five-, six-, and seven-membered benzosultams were efficiently assembled in an atom- and resource-economic manner. The generality of the process was demonstrated by forming cyclic products of various sizes from 42 substrates with different electronic effects and steric hindrance.

Annulative Morita-Baylis-Hillman reaction to synthesise chiral dibenzocycloheptanes

Summary: An enantioselective ring-opening formal [3+2]-cycloaddition of spirovinylcyclopropyl oxindoles with enals has been developed using synergistic catalysis of palladium(0) and a chiral organocatalyst, resulting in spirooxindoles bearing four contiguous stereocenters in good yields with excellent enantioselectivities. The protocol's generality and utility were confirmed through scale-up experiments and synthetic transformation of the resulting cycloadduct.
