Halogen-bond-assisted radical activation of glycosyl donors enables mild and stereoconvergent 1,2-cis-glycosylation

Summary: The chemistry of carbohydrates has a long history, and a simple, stereoselective and efficient glycosylation method is still needed for studying sugars. This study presents a strategy for 1,2-cis-glycosylation without using metals or strong acids. The method activates glycosyl donors through a radical cascade, resulting in the formation of 1,2-cis-glycosides with high stereoselectivity. The reaction shows remarkable tolerance to functional groups and is applicable to a broad range of donors.

Chemical synthesis of polysaccharides

Summary: In this article, we summarize the chemical syntheses of glycans containing over 20 monosaccharide units. The recent syntheses of ultra-long glycans, such as a 92mer arabinogalactan, a 100mer and a 151mer mannan, and a 128mer rhamnomannan, are highlighted. The assembly strategies, glycosylation methods, and protecting group manipulations, which are crucial for successful synthesis, are discussed to provide guidance for the future synthesis of various polysaccharides.

Total synthesis of Lentinus giganteus glycans with antitumor activities via stereoselective α-glycosylation and orthogonal one-pot glycosylation strategies

Summary: This paper reports an efficient, stereoselective and orthogonal one-pot synthesis of a tetradecasaccharide and shorter sequences from Lentinus giganteus polysaccharides. The method utilizes newly developed merging reagent modulation and remote anchimeric assistance for highly stereoselective glycosylation reactions. This method has mild reaction conditions, broad substrate scopes and significantly shortened step counts.

Leveraging Trifluoromethylated Benzyl Groups toward the Highly 1,2-Cis-Selective Glucosylation of Reactive Alcohols

Summary: By substituting the benzyl groups of glucosyl imidate donors with trifluoromethyl, a substantial increase in 1,2-cis-selectivity can be achieved when activated with TMS-I in the presence of triphenylphosphine oxide. The stereoselectivity depends on the number of trifluoromethyl groups, leading to high 1,2-cis-selectivity with reactive alcohol acceptors.

A novel one-pot glycosylation strategy based on glycosyl ortho-(1-phenylvinyl)benzoates was developed, enabling efficient synthesis of both glycans and nucleosides with expanded scope and leaving group options. This strategy was successfully applied to the total synthesis of capuramycin and divergent synthesis of TMG-chitotriomycin, offering a straightforward route to these biologically important molecules.
