Catalyst-Controlled Regiodivergent C-H Alkynylation of Thiophenes**

Arup Mondal et al.

Summary: Alkynes are important motifs in organic synthesis due to their presence in natural products and bioactive molecules. The method of inserting alkynes into (hetero)arenes relies heavily on the regioselectivity of the halogenation step, and can achieve selective alkynylation of thiophenes. The developed palladium-catalyzed C-H activation/alkynylation of thiophenes allows for regiodivergent reactions and late-stage modifications.

Transition-Metal-Catalyzed Divergent C-H Functionalization of Five-Membered Heteroarenes

Siyeon Jeong et al.

Summary: The regiodivergent, non-directed C-H functionalization of five-membered heteroarenes with alkenes and alkynes has been successfully achieved, allowing systematic diversification of common structural cores. A carefully devised transition metal catalytic system and a judicious choice of metals, ligands, acid and base additives, and solvents are critical for achieving high selectivity in these reactions.

Au-Ag Bimetallic Catalysis: 3-Alkynyl Benzofurans from Phenols via Tandem C-H Alkynylation/Oxy-Alkynylation

Long Hu et al.

Summary: This study presents a dual catalytic cycle for the synthesis of 3-alkynyl benzofurans, merging C-H alkynylation of phenols and oxy-alkynylation of triple bonds using the redox property and carbophilic pi acidity of gold. Mechanistic studies suggest the involvement of a bimetallic gold-silver species. The one-pot protocol allows for a direct, simple, and regio-specific approach with excellent functional group tolerance, making it suitable for the late-stage incorporation of benzofurans.

Double 1,2-Migration of Bromine and Silicon in Directed C-H Alkynylation Reactions with Silyl-Substituted Alkynyl Bromides through an Iridium Vinylidene Intermediate

Ken Yamazaki et al.

Summary: The recently developed iridium(III)-catalyzed C-H alkynylation provides an efficient strategy for constructing C(sp)-C(sp(2)) and C(sp)-C(sp(3)) bonds, with the mechanism extensively studied using density functional theory (DFT) calculations. The reactivity differences in the reaction can be attributed to the stabilization of the transition state by the beta-silicon effect in the 1,2-migration step.
