Recent Advances in Cross-Couplings of Functionalized Organozinc Reagents

Summary: Cross-couplings involving organozinc reagents can proceed well in the absence of transition-metal catalysts with reactive electrophiles. Highly diastereoselective and enantioselective cross-couplings can be readily performed with room-temperature configurationally stable alkylzinc species, producing diastereoselectively and enantiomerically enriched products. Highly regioselective magnesiations of functionalized arenes and heteroarenes undergo Negishi or Cu-catalyzed cross-couplings.

Photo and copper dual catalysis for allene syntheses from propargylic derivatives via one-electron process

Transition-Metal-Free Synthesis of Polyfunctional Triarylmethanes and 1,1-Diarylalkanes by Sequential Cross-Coupling of Benzal Diacetates with Organozinc Reagents

Summary: A variety of functionalized triarylmethane and 1,1-diarylalkane derivatives were successfully synthesized via a transition-metal-free, one-pot, two-step procedure, demonstrating the synthetic utility of the method for biologically relevant molecules. The sequential cross-coupling enabled by changing the solvent from THF to toluene and the proposed S(N)1-type mechanism were supported by experimental studies. The method was further demonstrated by the synthesis of several biologically relevant molecules, including an anti-tuberculosis agent, an anti-breast cancer agent, a precursor of a sphingosine-1-phosphate (S1P) receptor modulator, and a FLAP inhibitor.

Synthesis of Multisubstituted Allenes via Nickel-Catalyzed Cross-Electrophile Coupling

Summary: In this study, a cross-electrophile strategy was successfully applied to synthesize multisubstituted allenes. Nickel-catalyzed reductive cross-coupling between propargyl carbonates and organoiodides provided a method to prepare tri- or tetrasubstituted allenes without the need for pregenerated organometallics. Furthermore, propargyl carbonates were found to be suitable allenylating agents in nickel-catalyzed asymmetric reductive aryl-allenylation, leading to the formation of chiral benzene-fused cyclic compounds with a quaternary allenyl-substituted stereogenic center in a highly enantioselective manner.

Site-Selective Csp3-Csp/Csp3-Csp2 Cross-Coupling Reactions Using Frustrated Lewis Pairs

Summary: The donor-acceptor ability of frustrated Lewis pairs has been widely used in organic synthesis. Single electron transfer can generate a frustrated radical pair, leading to different reaction pathways and selectivity depending on the substrate and energy required. Investigation using EPR studies, kinetic studies, and DFT calculations has helped elucidate the mechanism of these coupling reactions and explain solvent-dependent site selectivity.

Enantioconvergent and Regioselective Synthesis of Allenylsilanes by Nickel-Catalyzed C(sp2)-C(sp3) Cross-Coupling Starting from Racemic a-Silylated Propargylic Bromides

Summary: This study reports a direct synthesis of enantioenriched allenylsilanes from racemic a-silylated propargylic bromides through an enantioconvergent nickel-catalyzed cross-coupling. The regioselectivity is controlled by the bulky silyl group, and the bond formation occurs exclusively at the y-position of the propargyl electrophile. The level of enantioselection induced by a chiral Pybox ligand is moderate.

Iron-Catalyzed Cross-Coupling of Propargyl Ethers with Grignard Reagents for the Synthesis of Functionalized Allenes and Allenols

Summary: An iron-catalyzed cross-coupling reaction of propargyl ethers with Grignard reagents was disclosed, allowing for efficient chirality transfer and preparation of various fluoroalkyl allenes. Additionally, an iron-catalyzed cross-coupling of Grignard reagents with alpha-alkynyl oxetanes and tetrahydrofurans was demonstrated as a straightforward approach towards fully substituted beta- or gamma-allenols.

The key to success lies in the design of chiral ligands to initiate the reaction and achieve enantiocontrol over the highly reactive prochiral alkyl radical species.
