Tubular dentin formation by TGF-β/BMP signaling in dental epithelial cells

Yoon Seon Lee et al.

Summary: This study identified that the Smad4-dependent TGF-beta 1/BMP2 signaling pathway regulates the expression of Cpne7 in dental epithelial cells, and the CPNE7 protein mediates odontoblast differentiation through epithelial-mesenchymal interaction. The results showed that conditioned medium treated with TGF-beta 1/BMP2 can induce the formation of tubular dentin.

The Genes Involved in Dentinogenesis

Shuang Chen et al.

Summary: The development and repair of dentin are regulated by hundreds of genes, and abnormalities are caused by gene mutations. Understanding this signal network is significant for tooth development, tissue regeneration, aging, and dental disease treatment, with molecular biology of genes as important future research basis.

The Modified Shields Classification and 12 Families with Defined DSPP Mutations

James P. Simmer et al.

Summary: Mutations in the DSPP gene can cause different types of dentin disorders, and this study identified disease-causing mutations and proposed a revised classification system based on the type of mutation rather than the severity of the phenotype. The study also revealed distinct cellular pathology differences between different DSPP mutations.

Oral manifestations of Ehlers-Danlos syndromes

Ulrike Lepperdinger et al.

Summary: Ehlers-Danlos syndromes (EDS) are inherited connective tissue disorders that manifest distinct pathologies in the teeth and oral cavity, affecting oral health-related quality of life. Standardization of diagnostic criteria is essential for resolving remaining questions, with specialized centers needing to apply congruent approaches to characterize clinical features and conduct genetic validation uniformly.

VPS4B deficiency causes early embryonic lethality and induces signal transduction disorders of cell endocytosis

Danna Chen et al.

Summary: Research has shown that VPS4B is involved in dentin dysplasia type I and plays a crucial role in tooth development. Deletion of VPS4B results in early embryonic lethality in mice, potentially due to disruptions in cell endocytosis signal transduction.

Summary: Periodontal Ehlers-Danlos syndrome (pEDS) is a rare condition caused by pathogenic variants in the C1R and C1S genes, which may present with early-onset periodontitis, skin issues, and vascular complications. Further research with larger case series is needed to enhance our understanding of the pathophysiology of this syndrome.

FGF23: A Review of Its Role in Mineral Metabolism and Renal and Cardiovascular Disease

Anna Kurpas et al.

Summary: FGF23, mainly secreted by osteocytes and osteoblasts, plays a crucial role in mineral ion homeostasis. Its relationship with phosphate and vitamin D metabolisms may lead to hereditary diseases. Elevated levels of FGF23 in early CKD stages and its association with cardiac complications have been observed.

Mariana del Pino et al.

Summary: This study provides clinical and radiological follow-up of four ODCD patients, with identification of four TRIP11 variants, two of which are novel. Most ODCD patients are compound heterozygotes for TRIP11 variants, although there is also one individual who is homozygous for a splice variant.

Biomineralization of Enamel and Dentin Mediated by Matrix Proteins

J. Moradian-Oldak et al.

Summary: Biomineralization of enamel, dentin, and bone involves the deposition of apatite mineral crystals within an organic matrix. While the structure and organic composition of these tissues differ, the molecular mechanisms involving protein interactions and crystallization events are common. The review highlights the importance of protein matrix control in the mineralization process and discusses strategies for biomimetic growth of enamel and repair of demineralized dentin and bone tissue engineering.

DSPP dosage affects tooth development and dentin mineralization

Dandrich Lim et al.

Summary: Dentin Sialoprotein (DSP) and phosphophoryn (PP) are two major non-collagenous proteins in dentin that play essential roles in dentin formation. Using transgenic technology, the DSPP gene knockout defect in mice can be partially or completely rescued, indicating the importance of DSPP in tooth development.

Mari Deguchi et al.

Summary: Osteogenesis imperfecta is a heterogeneous disorder primarily caused by genetic mutations, potentially fatal in severe cases, where prenatal examinations and genetic testing can aid in diagnosis.
