IMPDH dysregulation in disease: a mini review

Summary: Inosine-5'-monophosphate dehydrogenase (IMPDH) is an important enzyme in purine metabolism, regulating flux at the branch point between adenine and guanine nucleotide synthesis and participating in transcription regulation. Mutations in IMPDH1 and IMPDH2 are associated with different human diseases, but their effects at the enzyme level are still unclear. Mutations in IMPDH1 lead to retinal degeneration and recent studies have revealed the molecular mechanisms behind the functional defects. IMPDH1 splice variants in the retina up-regulate guanine nucleotide synthesis by reducing sensitivity to feedback inhibition by downstream products. Understanding the role of IMPDH1 in the retina and characterizing an animal disease model will provide insights into the molecular mechanism of IMPDH1-associated blindness.

A highly conserved zebrafish IMPDH retinal isoform produces the majority of guanine and forms dynamic protein filaments in photoreceptor cells

Summary: In this study, Impdh1a in the zebrafish retina was investigated. It was found that Impdh1a plays a significant role in rod and cone photoreceptors and its loss leads to a reduction in guanine levels without affecting cellular morphology and cGMP levels. These findings provide fundamental new insights into the function of Impdh1a in the zebrafish retina.

Electrophysiology-Guided Genetic Characterisation Maximises Molecular Diagnosis in an Irish Paediatric Inherited Retinal Degeneration Population

Summary: Inherited retinal degenerations (IRDs) account for a significant proportion of pediatric blindness, and there are often long delays in reaching a diagnosis. In this study, a telegenetics approach was used to offer genetic testing to children with phenotypic evidence of IRD during the COVID-19 pandemic. Genetic testing identified causative genetic variants in a high percentage of patients, and a significant proportion were eligible for approved therapies or clinical trial-based gene therapies. Early introduction of genetic testing is crucial for genetic counseling and preparation for upcoming gene therapy trials in pediatric IRD patients.

Multimodal imaging of retinitis pigmentosa associated with Mainzer-Saldino syndrome

Despite carrying the same mutations and severe retinitis pigmentosa phenotype, the younger sister showed slightly more advanced retinal degeneration.
