Frameshifting at collided ribosomes is modulated by elongation factor eEF3 and by integrated stress response regulators Gcn1 and Gcn20

Lisa Houston et al.

Summary: This study reveals the factors involved in frameshifting during ribosome collisions and finds that eEF3, Gcn1, and Gcn20 have opposing roles in modulating frameshifting. The results show that eEF3 facilitates tRNA-mRNA translocation and E-site tRNA release, thereby suppressing frameshifting; while Gcn1 and Gcn20 bind to collided ribosomes and increase frameshifting. Additionally, Mbf1 requires He12 or Gcn1 to suppress frameshifting.

Redox-sensitive E2 Rad6 controls cellular response to oxidative stress via K63-linked ubiquitination of ribosomes

Vanessa Simoes et al.

Summary: Protein ubiquitination is an important process that regulates protein synthesis, function, and fate. The E2 Rad6 associates and modifies ribosomes during stress, and its activity is redox regulated. This study provides insights into the role and regulation of E2 enzymes in the ubiquitination pathway and reveals the physiological role of Rad6 in translation, antioxidant defense, and stress adaptation.

Frameshifting at collided ribosomes is modulated by elongation factor eEF3 and by integrated stress response regulators Gcn1 and Gcn20

Lisa Houston et al.

Summary: Research has shown that in Saccharomyces cerevisiae, the translation elongation factor eEF3 and the integrated stress response (ISR) pathway components Gcn1 and Gcn20 modulate frameshifting in opposing manners. eEF3 facilitates tRNA-mRNA translocation and E-site tRNA release, suppressing frameshifting, while removal of Gcn1 and Gcn20 increases frameshifting.

The La-related proteins: structures and interactions of a versatile superfamily of RNA-binding proteins

Anne-Catherine Dock-Bregeon et al.

Summary: The La-related proteins (LaRPs) are an ancient superfamily of RNA-binding proteins that orchestrate the main fates of RNA through a complex network of protein-RNA and protein-protein interactions. Recent studies have revealed that LaRPs have evolved unique mechanisms of RNA recognition involving structured domains and disordered regions, working together to achieve RNA substrate discrimination. This review highlights our current understanding of the structure-function relationship within the LaRP superfamily.

Controversies around the function of LARP1

Andrea J. Berman et al.

Summary: LARP1 has attracted attention for its involvement in the mTOR signaling cascade and regulation of TOP mRNA translation. However, there are conflicting findings on whether LARP1 represses or activates TOP translation. In this study, opinions from four leading scientists in the field are presented to discuss these contradictory findings.

Ribosome quality control antagonizes the activation of the integrated stress response on colliding ribosomes

Liewei L. Yan et al.

Summary: Translation stalling activates ribosome quality control (RQC) and is linked to the integrated stress response (ISR) through Gcn2, with Hel2 suppressing Gcn2 activation. Both pathways are activated by similar factors causing ribosome stalling, although Hel2 is maximally activated at a lower frequency. Inactivation of one pathway leads to overactivation of the other, suggesting they are both triggered by the same signal. ISR prefers an empty A site while RQC shows no preference, indicating they do not directly compete with each other.

Ribosome states signal RNA quality control

Karole N. D'Orazio et al.

Summary: Eukaryotic cells integrate multiple quality control responses during protein synthesis, signaled by slow or stalled elongating ribosomes. Depending on the nature of the delay, the signal may lead to translational repression, messenger RNA decay, ribosome rescue, and/or nascent protein degradation. The structure and composition of an elongating ribosome in a troubled state determine the downstream quality control pathways involved, highlighting the intersecting pathways between RNA decay and ribosome rescue.

Structure of Gcn1 bound to stalled and colliding 80S ribosomes

Agnieszka A. Pochopien et al.

Summary: The Gcn pathway is highly conserved in eukaryotes, with Gcn1 serving as an essential effector protein for Gcn2 kinase to regulate global translation in response to stress. The structure of Gcn1 interacting with stalled and colliding ribosomes was revealed through cryo-electron microscopy, shedding light on the activation of Gcn2. This study provides important insights into the molecular mechanism of Gcn pathway activation and its implications in ribosome-associated quality control pathways.

The ribosome collision sensor Hel2 functions as preventive quality control in the secretory pathway

Yoshitaka Matsuo et al.

Summary: The study finds that Hel2 preferentially binds to pre-engaged secretory ribosome-nascent chain complexes, especially under signal recognition particle (SRP) deficient conditions. Deletion of Hel2 exacerbates mitochondrial defects caused by insufficient SRP, leading to mistargeting of secretory proteins into mitochondria. These insights shed light on risk management in the secretory pathway for maintaining cellular protein homeostasis.
