Life histories of myeloproliferative neoplasms inferred from phylogenies

Nicholas Williams et al.

Summary: By sequencing the clones of 12 patients with myeloproliferative neoplasms, we identified that early driver mutations are the foundation for tumor development and are linked to lifelong growth and evolution. This discovery provides opportunities for early intervention and a new model for cancer development.

All2: A tool for selecting mosaic mutations from comprehensive multi-cell comparisons

Vivekananda Sarangi et al.

Summary: Mosaic mutations play a vital role in understanding normal development and diseases. We have developed a novel method, All(2), that accurately detects mosaic mutations by comparing the genomes of single cells. This method performs well in high-resolution cell lineage tracing and reduces false positives.

Somatic mutation landscapes at single-molecule resolution

Federico Abascal et al.

Summary: The study developed a method to detect mutations in single cells or small clones, revealing that somatic mutations also occur in non-dividing cells and contribute significantly to somatic mutagenesis.

Lineage tracing of human development through somatic mutations

Michael Spencer Chapman et al.

Summary: By reconstructing a phylogenetic tree of blood development in healthy human fetuses, it was found that individual haematopoietic progenitors acquire multiple somatic mutations by 18 weeks after conception. The data also support a hypoblast origin of the extra-embryonic mesoderm and primitive blood in humans.

Early developmental asymmetries in cell lineage trees in living individuals

Liana Fasching et al.

Summary: The study found that mosaic mutations can help track cell lineages in humans, with different efficiencies of DNA repair potentially contributing to lineage imbalances. By analyzing cell lineages, insights into cell division, tissue formation, and disease development can be gained.

Extensive phylogenies of human development inferred from somatic mutations

Tim H. H. Coorens et al.

Summary: This study reconstructed phylogenies of normal tissues from three adult individuals using whole-genome sequencing, revealing asymmetries in cell contributions and spatial embryonic patterning within and between individuals. The research also showed a spatial effect in brain development and suggested extraembryonic contributions to primordial germ cells. Despite reaching the same ultimate tissue patterns, early bottlenecks and lineage commitments lead to substantial variation in embryonic patterns.

Clonal dynamics in early human embryogenesis inferred from somatic mutation

Seongyeol Park et al.

Summary: This study used genomic analysis of human embryos and adult tissues to reconstruct cellular phylogenies, revealing unequal contribution of early cells to embryo development and examples of clonal imbalances between tissues. The research also provided insights into age-related mutational processes and loss of sex chromosomes in normal somatic cells.
