A conserved arginine in NS5 binds genomic 3′ stem-loop RNA for primer-independent initiation of flavivirus RNA replication

Sai Wang et al.

Summary: This study reveals the interaction between the nonstructural protein NS5 and the stem-loop A (SLA) in the 5' UTR, as well as the top-loop of 3' SL in the 3' UTR during flavivirus RNA replication. The conserved arginine 888 of DENV2 NS5 plays a crucial role in virus viability and de novo initiation of RNA polymerase activity.

Dynamic bulge nucleotides in the KSHV PAN ENE triple helix provide a unique binding platform for small molecule ligands

Monalisa Swain et al.

Summary: Cellular and virus-coded long non-coding RNAs play multiple roles in biological and pathological processes, with some evading degradation machinery to support disease progression. A triplex structure involving the 3' terminus of lncRNAs and nuclear expression element stabilizes RNA transcripts, presenting a potential therapeutic target. Screening of small molecules identified compounds binding to this triplex structure, with crystal structures revealing local structural rearrangements and molecular dynamics simulations corroborating the ligand binding role of a dinucleotide bulge. This study supports the potential of targeting RNA triplexes with small molecules.

Structures of flavivirus RNA promoters suggest two binding modes with NS5 polymerase

Eunhye Lee et al.

Summary: Flaviviruses use stem-loop A (SLA) as a promoter for RNA synthesis, with NS5 recognizing SLA to initiate RNA synthesis. The crystal structures of DENV and ZIKV SLAs show differences in orientation, but these can be compensated by differences in domain arrangements in DENV and ZIKV NS5s.

Establishing RNA-RNA interactions remodels lncRNA structure and promotes PRC2 activity

Maggie M. Balas et al.

Summary: This study reveals that intermolecular RNA-RNA interactions between long noncoding RNAs and their targets can relieve the inhibitory effect of PRC2. RNA bridging is promoted by heterogeneous nuclear ribonucleoprotein B1. Additionally, establishing RNA-RNA interactions can change the structure of lncRNA HOTAIR.
