Photoactivatable V-Shaped Bifunctional Quinone Methide Precursors as a New Class of Selective G-quadruplex Alkylating Agents

Alberto Lena et al.

Summary: The combination of G4 ligands and photochemistry provides a new strategy to study the biological relevance of G4 structures. This study developed six novel V-shaped G4 ligands that can form stable covalent adducts with G4 structures through photochemical reactions. The ligands exhibit preferential binding towards the human telomeric sequence.

Advances in covalent drug discovery

Lydia Boike et al.

Summary: Covalent drugs have been widely used for over a century, but recent advances in tools for rational design have led to the successful development of potent and selective inhibitors, such as KRAS(G12C) inhibitors. The use of electrophile-first approaches and the emergence of chemoproteomics platforms have greatly enhanced the discovery and development of covalent drugs.

Synthesis and structure-activity relationship of peptide nucleic acid probes with improved interstrand-crosslinking abilities: application to biotin-mediated RNA-pulldown

Enrico Cadoni et al.

Summary: The development of interstrand-crosslinking probes for DNA and RNA has been extensively studied, but most reactions cannot be reversed. This study reports the reversibility of furan-mediated interstrand-crosslinking reaction and demonstrates the benefits of using this method in cell lysates.

New (Iso)quinolinyl-pyridine-2,6-dicarboxamide G-Quadruplex Stabilizers. A Structure-Activity Relationship Study

Enrico Cadoni et al.

Summary: The study aims to synthesize analogues of G4 ligand 360A, identify structure-activity relationships, and design other G4-interactive small molecules. Experimental results show that methylation affects the stability and selectivity of compounds towards G4, with compounds having a relative 1,3-position displaying the best G4 stabilization.

Beyond small molecules: targeting G-quadruplex structures with oligonucleotides and their analogues

Enrico Cadoni et al.

Summary: G-Quadruplexes (G4s) are secondary structures of DNA/RNA that naturally occur in genome regions like proto-oncogenes and telomeres, influencing gene expression regulation and other biological processes. While many small molecules capable of stabilizing G4s have been reported in the past two decades, approaches based on synthetic probes for sequence-specific targeting are still limited.

Summary: PNA, with its unique properties, is a promising candidate for therapeutic and biotechnological interventions, particularly in gene expression regulation and gene editing. While traditional PNA design has been widely used, advancements in technology can be made through chemical modifications of the PNA backbone and nucleobases. This review discusses recent developments in PNA-based gene manipulation techniques and the use of novel chemical modifications to enhance PNA-mediated gene targeting.

Teaching photosensitizers a new trick: red light-triggered G-quadruplex alkylation by ligand co-localization

Enrico Cadoni et al.

Summary: A novel bimolecular approach for G-quadruplex alkylation using a pro-reactive furan-containing ligand activated by red-light irradiation of a G4-binding photosensitizer is proposed. The method allows for selective alkylation of G4 over dsDNA and shows high yields at low ligand excess. HPLC and modeling studies help identify potential residues involved in alkylation.
