Trauma-focused cognitive-behavioral therapies for posttraumatic stress disorder under ongoing threat: A systematic review

Naomi Ennis et al.

Summary: This paper systematically reviewed the effectiveness of CBTs for PTSD in individuals with ongoing threat of reexposure and found medium to large effects from pre to posttreatment. However, there were mixed findings for long-term outcomes in domestic violence samples. Treatment adaptations focused on establishing relative safety and differentiating between realistic threat and generalized fear responses. Further research is needed to draw firm conclusions on the safety and effectiveness of trauma-focused CBTs for individuals under ongoing threat.

Limbic Neuropeptidergic Modulators of Emotion and Their Therapeutic Potential for Anxiety and Post-Traumatic Stress Disorder

Paul J. Marvar et al.

Summary: Post-traumatic stress disorder (PTSD) is associated with dysregulation of limbic neuropeptides, with studies showing that blocking certain pathways can weaken fear memory consolidation, enhance fear extinction, and improve threat discrimination. Novel approaches targeting these pathways offer promising potential for treating core elements of PTSD pathophysiology.

Localization and expression of the Mas-related G-protein coupled receptor member D (MrgD) in the mouse brain

Javad Hami et al.

Summary: Numerous studies have shown evidence of a local renin-angiotensin system (RAS) in the central nervous system (CNS), with the identification of a novel receptor known as Mas-related G-protein coupled receptor D (MrgD). The distribution pattern of MrgD receptors in the adult mouse brain reveals its presence in specific areas related to reward and limbic functions, suggesting a potential role in processes such as pain perception/modulation, synaptic plasticity, learning, memory, and cognition.

Central amygdala micro-circuits mediate fear extinction

Nigel Whittle et al.

Summary: Fear extinction involves the reduction of defensive responses following repeated exposure to fear-related stimuli without harm, with recent studies indicating the involvement of central amygdala inhibitory microcircuits in this process. These microcircuits produce reversible, stimulus- and context-specific changes in neuronal responses, absent in cases of deficient extinction, and impair fear extinction when PKC delta neurons are selectively silenced.
