Neural mechanism underlying depressive-like state associated with social status loss

Zhengxiao Fan et al.

Summary: This study demonstrates that downward social mobility and depressive behaviors are interconnected through neural mechanisms involving negative reward prediction error and the activation of the lateral habenula (LHb). Forced loss in a non-violent social contest generates negative reward prediction error, leading to the activation of LHb and inhibition of the medial prefrontal cortex (mPFC), resulting in retreats in contests. These findings provide insights into the relationship between social status loss and depression, and offer a potential target for intervention.

Dual action of ketamine confines addiction liability

Linda D. Simmler et al.

Summary: Ketamine does not induce synaptic plasticity in the nucleus accumbens despite eliciting dopamine transients, due to the rapid off-kinetics of the dopamine transients and NMDAR antagonism. However, it supports reinforcement through NMDAR antagonism in GABA neurons of the ventral tegmental area, which is quickly terminated by type-2 dopamine receptors on dopamine neurons.

A non-hallucinogenic psychedelic analogue with therapeutic potential

Lindsay P. Cameron et al.

Summary: The study identifies key structural elements of the potential therapeutic pharmacophore of ibogaine and designs tabernanthalog, a safer, non-hallucinogenic analogue of ibogaine. Tabernanthalog is found to promote neural plasticity, reduce addiction-related behavior, and produce antidepressant effects in rodents. Through careful chemical design, it is possible to modify psychedelic compounds to produce safer, non-hallucinogenic variants with therapeutic potential.

Synaptic mechanism underlying serotonin modulation of transition to cocaine addiction

Yue Li et al.

Summary: This study found that cocaine use enhances compulsive behavior by affecting serotonin levels, while this effect can be reversed by increasing serotonin levels. The presynaptic depression induced by 5-HT via 5-HT1B receptors plays a role in the bidirectional effect on behavior.
