Host Tau Genotype Specifically Designs and Regulates Tau Seeding and Spreading and Host Tau Transformation Following Intrahippocampal Injection of Identical Tau AD Inoculum

Pol Andres-Benito et al.

Summary: This study focuses on the significance of host tau in tau seeding and spreading, as well as the molecular changes associated with the transformation of host tau into abnormal tau. The findings show that host tau is necessary for tau seeding and spreading following inoculation with sarkosyl-insoluble tau fractions from AD brains. The study also reveals that tau seeding is accompanied by genotype-dependent tau phosphorylation, tau nitration, MAP2 phosphorylation, and activation of specific kinases.

Transcriptional and Post-Transcriptional Regulation of Oxytocin and Vasopressin Gene Expression by CREB3L1 and CAPRIN2

Soledad Barez-Lopez et al.

Summary: This study explores the expression of two genes, Caprin2 and Creb3l1, in magnocellular neurones (MCNs) and their changes during dehydration. The findings suggest that both genes are expressed in MCNs and their expression increases in response to dehydration. Protein levels mirror the increase in transcript levels for both CREB3L1 and CAPRIN2. In addition, CREB3L1 may be a transcription factor regulating Oxt gene expression, while CAPRIN2 binds to Oxt mRNA and regulates its poly(A) tail length.

Short-Term Administration of Common Anesthetics Does Not Dramatically Change the Endogenous Peptide Profile in the Rat Pituitary

Somayeh Mousavi et al.

Summary: Cell-cell signaling peptides play important roles in various physiological processes, and liquid chromatography-mass spectrometry-based peptidomics workflows are commonly used to identify and quantify their relative abundances. However, acute administration of anesthetics may affect the measured abundance of cell-cell signaling peptides, especially in the brain and endocrine system. This study evaluated the effect of short-term anesthetic administration on the measured abundance of these peptides and found that isoflurane had a minor impact on the levels of some peptides, while sodium pentobarbital had negligible impact.

Summary: This article reviews the impact of weak magnetic fields on biological systems and proposes an explanation based on the radical pair mechanism. By studying static, hypomagnetic, and oscillating magnetic fields, as well as isotope effects, it is found that magnetosensitivity is widespread throughout biology.

Aquaporin 1 mediates early responses to osmotic stimuli in endothelial cells via the calmodulin pathway

Yong Jiang et al.

Summary: The study found that AQP1 responds to osmotic stimuli by redistributing to the cell membrane and influencing changes in cell size. Exposure to hypo- or hypertonic medium can induce alterations in cell volume, and calmodulin kinase appears to regulate AQP1 activity during the early response phase.

Prolonged anesthesia alters brain synaptic architecture

Michael Wenzel et al.

Summary: Prolonged medically induced coma (pMIC) is routinely carried out in intensive care medicine, leading to cognitive impairment. A study on adolescent and mature mice shows that pMIC affects object recognition and is associated with enhanced synaptic turnover, with increased synapse formation during pMIC and synaptic loss post-anesthesia. These results demonstrate significant side effects of prolonged anesthesia on neural circuit structure.

Sensorimotor Perturbation Induces Late and Transient Molecular Synaptic Proteins Activation and Expression Changes

Julie Fourneau et al.

Summary: This study investigated the synaptic plasticity process in the somatosensory primary cortex during a period of sensorimotor perturbation and recovery. The results showed that changes in the presynaptic compartment occurred as early as D7 during SMP, followed by changes in the postsynaptic compartment at D14, and these changes persisted at least until 24 hours of recovery. Proteins related to synapse structure were mildly affected, primarily at D14.

Radical pairs may play a role in xenon-induced general anesthesia

Jordan Smith et al.

Summary: This study investigates the xenon-induced general anesthesia process and suggests that the potency of xenon as a general anesthetic is influenced by isotopic dependence. They propose a mechanism where xenon nuclear spin may affect the recombination dynamics of radical pairs of electrons. Their model is able to reproduce the observed isotopic dependence in the potency of xenon as a general anesthetic, supporting the idea that radical pairs of electrons with entangled spins could play a role in consciousness.

Dysregulated protein phosphorylation: A determining condition in the continuum of brain aging and Alzheimer's disease

Isidro Ferrer et al.

Summary: The study found abnormal protein phosphorylation at the early stages of NFT pathology in the elderly and sporadic Alzheimer's disease (sAD), with protein phosphorylation abnormalities in the FC preceding the formation of NFTs and SPs. The most active period of dysregulated phosphorylation is at stages III-IV, when some individuals may be classified clinically as having mild cognitive impairment, which is a precursor stage in the progression to dementia.

Dendritic spine remodeling and plasticity under general anesthesia

Simon Granak et al.

Summary: General anesthesia, considered a medical miracle, enables life-saving interventions without pain sensation and traumatic memories. However, the mechanisms of how it induces a coma-like state are not fully understood. Research shows that even brief exposure to general anesthesia can have lasting effects on brain plasticity, cognition, mood, and social behavior.

The multiple faces of the oxytocin and vasopressin systems in the brain

Valery Grinevich et al.

Summary: Traditionally, hypothalamic neuroendocrine cells were classified into magnocellular and parvocellular neurons for hormone release and brain circuit regulation purposes respectively. However, recent studies show these classic projections are outdated, with oxytocin and vasopressin released in the brain from dendrites and distant axon varicosities to modulate information transfer through conventional synapses.

Anesthesia in mice activates discrete populations of neurons throughout the brain

Shai-Lee Yatziv et al.

Summary: The brain undergoes rapid, dramatic, and reversible transitions between wakefulness and unconsciousness during sleep and in pathological conditions. Selective transitioning allows for the persistence of basic functions while mobility, sensory perception, memory formation, and awareness are lost. The role of specific brainstem and diencephalic arousal nuclei in driving these transitions has been implicated.

Post-translational modifications in proteins: resources, tools and prediction methods

Shahin Ramazi et al.

Summary: Posttranslational modifications (PTMs) involve modifications to amino acid side chains in proteins after biosynthesis, affecting various aspects of protein functions. Disruptions in PTMs can lead to diseases, emphasizing the need for computational methods to predict PTMs. High-throughput experimental methods for PTM discovery are laborious, prompting the exploration of computational tools and databases to advance research in this area.
