YTHDF1 promotes mRNA degradation via YTHDF1-AGO2 interaction and phase separation

Jiong Li et al.

Summary: YTHDF1 recruits AGO2 through the YTH domain and degrades target mRNAs by promoting P-body formation through LLPS. Deletion of YTHDF1 causes P-bodies to transition from liquid droplets to gel/solid droplets, forming AGO2/RNA patches and delaying mRNA degradation. These findings indicate a previously unrecognized crosstalk between YTHDF1 and AGO2 in post-transcriptional mRNA regulation.

LncRNAs divide and rule: The master regulators of phase separation

Kumaravel Somasundaram et al.

Summary: Most of the human genome codes for non-protein-coding RNAs, specifically long non-coding RNAs (lncRNAs), which play diverse roles in physiological and pathological processes. The formation of membrane-less organelles (MLOs) through liquid-liquid phase separation (LLPS) is crucial for carrying out specialized tasks inside the cell. While the role of RNA binding proteins (RBPs) in this process is known, the specific contribution of RNAs is not well-understood. This review aims to update the current understanding of LLPS, the involvement of lncRNAs, and its relevance to cancer development and progression.

Summary: Liquid-liquid phase separation (LLPS) is a phenomenon that has attracted significant attention in biological studies. However, the comprehensive understanding of its physical mechanism is limited. The characteristics of side chains of amino acids and the interaction tendency of proteins play important roles in regulating LLPS and should receive more attention. This review introduces the driving forces and protein secondary structures involved in LLPS, summarizes existing reports on post-translational modifications (PTMs) related to LLPS, and analyzes the underlying basic principles to find potential connections between LLPS and PTM.

RNA contributions to the form and function of biomolecular condensates

Christine Roden et al.

Summary: Recent studies have shown the significant contribution of RNA to cellular liquid-liquid phase separation and condensate formation, which play important roles in various cellular functions such as RNA transport, supporting catalytic processes, and stress responses. Although RNA and RNA-protein condensates are important in cells, the role of RNA in liquid-liquid phase separation has received less attention in comparison to RNA-binding proteins.Properties such as composition, length, structure, modifications, and expression levels of RNA can modulate the biophysical features of native condensates, influencing their size, shape, viscosity, and composition. RNA-protein condensates are involved in various cellular functions, including cell compartmentalization through RNA transport and localization, supporting catalytic processes, storage and inheritance of specific molecules, as well as buffering noise and responding to stress.

Contrasting Effects of Ferric and Ferrous Ions on Oligomerization and Droplet Formation of Tau: Implications in Tauopathies and Neurodegeneration

Sandipan Mukherjee et al.

Summary: The dysregulation of ferric ions leads to enhanced aggregation of tau protein, with Fe3+ decreasing the critical concentration required for liquid-liquid phase separation of tau and significantly increasing the formation of tau oligomers and fibrils. Fe3+ binds to tau with higher affinity compared to Fe2+ and disrupts the secondary structure of tau, favoring tau aggregation by increasing beta-sheet content and decreasing disordered content. Fe3+ may enhance non-covalent interactions between disordered domains of tau molecules, ultimately leading to tau aggregation and pointing to a potential relationship between dysregulation of ferric ions and neurodegenerative disorders.

Phase separation of p53 precedes aggregation and is affected by oncogenic mutations and ligands

Elaine C. Petronilho et al.

Summary: Mutant p53 tends to form aggregates with amyloid properties, especially amyloid oligomers inside the nucleus, which are believed to cause oncogenic gain-of-function. The DNA-binding domain of p53 (p53C) undergoes phase separation and aggregation in the presence of polyethylene glycol (PEG), and mutant p53C undergoes phase separation faster than wild-type p53C. Mutant p53 undergoes gel- and solid-like phase transitions in the nucleus, modulated by polyanions like heparin and RNA. The disordered transactivation domain (TAD) also plays a role in phase separation and amyloid aggregation of p53.
