High-throughput and high-content bioassay enables tuning of polyester nanoparticles for cellular uptake, endosomal escape, and systemic in vivo delivery of mRNA

Yuan Rui et al.

Summary: Nanoparticle-based mRNA therapeutics show great promise, but cellular internalization and endosomal escape pose challenges for cytosolic delivery. Researchers developed a dual nanoparticle uptake and endosomal disruption assay, which can detect cellular internalization and endosomal disruption. The study found that this assay has higher predictive capacity for mRNA delivery compared to conventional polymer and nanoparticle characteristics.

CRISPR activation screen identifies BCL-2 proteins and B3GNT2 as drivers of cancer resistance to T cell-mediated cytotoxicity

Julia Joung et al.

Summary: This study identifies several genes associated with tumor resistance to T cell cytotoxicity through a genome-scale CRISPR activation screen. Overexpression of these candidate genes, including MCL1, JUNB, and B3GNT2, confers resistance to T cell killing in various cancer cell types and mouse xenografts. Mechanistically, MCL1 and JUNB modulate the mitochondrial apoptosis pathway, while B3GNT2 disrupts interactions between tumor and T cells, leading to reduced T cell activation. Inhibition of these candidate genes sensitizes tumor models to T cell cytotoxicity.

Adoptive cell therapy with tumor-specific Th9 cells induces viral mimicry to eliminate antigen-loss-variant tumor cells

Gang Xue et al.

Summary: This research demonstrates that tumor-specific Th9 cells have a unique capacity to eliminate advanced tumors containing ALVs, attributed to their enhanced direct tumor cell killing and interferon alpha/beta-induced bystander antitumor effects. This is due to the accumulation of extracellular ATP within the tumor microenvironment, promoting immune cell infiltration and antitumor cytokine production.

A metabolic switch to memory CAR T cells: Implications for cancer treatment

Hosein Rostamian et al.

Summary: The therapeutic efficacy of CAR T cells in hematological malignancies is remarkable, but limited in solid tumors due to challenges of persistence and dysfunction in an immunosuppressive tumor microenvironment. Reprogramming CAR T cell metabolism and targeting metabolic pathways can enhance antitumor activity, with strategies focusing on promoting longevity and altering cellular fate within tumors.

The metabolism of cancer cells during metastasis

Gabriele Bergers et al.

Summary: This review discusses the metabolic rewiring in cancer cells during the metastatic cascade and the distinct metabolic features of primary tumors and metastases. Metastasis is the leading cause of death in most cancer patients, and targeting metastatic seeding and colonization remains a significant challenge. Adjusting metabolism may provide therapeutic opportunities to prevent or reduce metastasis, and targeting the new metabolic traits in established metastases could lead to their eradication.
