Genetically programmed cell-based synthesis of non-natural peptide and depsipeptide macrocycles

Summary: We have successfully synthesized 25 diverse non-natural macrocyclic peptides and depsipeptides, each containing two non-canonical amino acids, through encoded synthesis. By modifying the genome, Syn61 & UDelta;3-derived cells were able to synthesize non-natural peptides, with the annotated occurrences of two sense codons and a stop codon, and the corresponding transfer RNA and release factor removed. We have also engineered distinct pyrrolysyl-tRNA synthetase/tRNA pairs to direct the incorporation of diverse alpha hydroxy acids.

Measuring the tolerance of the genetic code to altered codon size

Summary: This study investigates the tolerance of the Escherichia coli genetic code to tRNA mutations and found that while most tRNAs can be converted to a quadruplet form, only nine aminoacyl tRNA synthetases can tolerate quadruplet anticodons. This limits the chemical alphabet available for an all-quadruplet code.

Summary: The Escherichia coli tyrosyl-tRNA synthetase/tRNA pair has been widely used in genetic engineering in eukaryotes. By engineering the tRNA and developing an optimized selection system, the cross-reactivity issues with bacterial glutaminyl-tRNA synthetase were resolved, enabling the efficient enrichment of new amino acid-selective synthetase mutants in mammalian cells. The platform has proven to be valuable for incorporating noncanonical amino acids.

Summary: This study demonstrated the ribosomal elongation of linear and statines amino acids to expand the diversity of nonproteinogenic amino acids in combinatorial peptide libraries. The incorporation of statines into peptides could potentially confer important pharmacological properties, such as protease resistance and membrane permeability, and enhance the potential of future drug discovery.
