ATM-SPARK: A GFP phase separation-based activity reporter of ATM

Xiaoquan Li et al.

Summary: In this study, a robust ATM activity reporter called ATM-SPARK was designed based on phosphorylation-inducible green fluorescent protein phase separation. The reporter allows visualization of endogenous ATM activity and DNA damage in living cells, and can be used for high-throughput screening. The study identified protein phosphatase 4 as an ATM activity blocker and the compound BGT226 as a potent ATM inhibitor.

Force-FAK signaling coupling at individual focal adhesions coordinates mechanosensing and microtissue repair

Dennis W. Zhou et al.

Summary: The coupling of traction force and FAK signaling coordinates cell migration and tissue-scale forces to promote microtissue repair. This process relies on actomyosin contractility, talin-FAK binding, and vinculin to convert forces into FAK signaling events regulating cell migration and tissue repair.

Turn-key mapping of cell receptor force orientation and magnitude using a commercial structured illumination microscope

Aaron Blanchard et al.

Summary: The article introduces a nucleic acid-based molecular tension probe and molecular force microscopy (MFM) technology, which use fluorescence polarization to map the direction of pN forces applied by receptors. By utilizing structured illumination microscopy (SIM), super-resolution MFM is achieved, generating high-resolution maps of both the magnitude and orientation of pN traction forces applied by cells.

An Outside-In Switch in Integrin Signaling Caused by Chemical and Mechanical Signals in Reactive Astrocytes

Leonardo A. Perez et al.

Summary: Astrocyte reactivity is influenced by mechanical and chemical stimuli, with integrins playing a key role in triggering cell contraction and adhesion in response to Thy-1 engagement. This study highlights the potential of targeting alpha(V)beta(3) Integrin mechanoreceptor to regulate astrocyte behavior in neuronal healing.
