Nanosensor-based monitoring of autophagy-associated lysosomal acidification in vivo

Summary: Autophagy is a cellular process important for neurodegenerative diseases and cancers. However, current methods for measuring lysosomal pH, a crucial aspect of autophagy, lack quantification and in vivo capabilities. In this study, near-infrared optical nanosensors were developed to quantitatively measure autophagy-mediated endolysosomal hyperacidification in live cells and in vivo. The sensors localized to lysosomes and allowed dynamic and quantitative mapping of subtle changes in lysosomal pH. Using these sensors, cellular and intratumoral hyperacidification was observed, providing insights into the dynamics of autophagy-lysosomal pathway.

By developing a new zinc ion fluorescent probe and combining it with SIM imaging technology, the study successfully achieved morphology-correlated organelle identification in living cells.
