Metallaphotoredox: The Merger of Photoredox and Transition Metal Catalysis

Summary: The merger of photoredox catalysis with transition metal catalysis, known as metallaphotoredox catalysis, has become an important part of synthetic methodology. It combines the bond formation capability of transition metal catalysis with the utility of photoinduced electron and energy transfer processes. Photocatalytic substrate activation allows the involvement of simple starting materials in metal-mediated bond-forming processes, while electron or energy transfer with organometallic intermediates provides a complementary activation mode to traditional catalytic platforms.

Intermolecular [2π+2σ]-photocycloaddition enabled by triplet energy transfer

Summary: In this study, a new intermolecular [2+2]-photocycloaddition method using bicyclo[1.1.0]butanes as 2 sigma-electron reactants was reported. This reaction was realized by visible-light-mediated triplet energy transfer catalysis. The method allows for a simple, modular, and diastereoselective synthesis of bicyclo[2.1.1]hexanes and extends to a-bonds, providing previously inaccessible structural motifs.

Photochemistry of (η4-diene)Ru(CO)3Complexes as PrecursorCandidates for Photoassisted Chemical Vapor Deposition

Summary: Solution phase photochemical experiments showed that (eta 4-diene)-Ru(CO)3 complexes can be used as precursors for photoassisted chemical vapor deposition of ruthenium by losing both diene and CO ligands. The quantum yields of ligand loss were found to depend on the structure of the diene, and subsequent ligand loss was determined to occur from secondary photolysis. The compounds exhibited little luminescence, indicating that radiative decay of the excited states was not competitive with photochemical ligand loss. Additionally, the formation of colloidal Ru species was observed after irradiation in hydrocarbon solutions in the absence of phosphite.

Practical and Facile Access to Bicyclo[3.1.1]heptanes: Potent Bioisosteres of meta-Substituted Benzenes

Summary: There is growing interest in replacing the planar aromatic rings in drug candidates with three-dimensional caged scaffolds. However, finding bioisosteres of meta-substituted benzenes has been a challenge. In this study, the bicyclo[3.1.1]heptane scaffold was explored as a novel bioisostere, and a practical method for its synthesis was developed.

Synthesis of meta-substituted arene bioisosteres from [3.1.1]propellane

Summary: Small-ring cage hydrocarbons are popular bioisosteres for para-substituted benzene rings in drug design. In this study, we synthesized hydrocarbons called bicyclo[3.1.1]heptanes (BCHeps) from [3.1.1]propellane, which have geometric correspondence to meta-substituted benzenes. The BCHeps can be easily transformed into medicinally relevant carbon- and heteroatom-substituted compounds, displaying enhanced metabolic stability compared to their parent arene-containing drugs. This research demonstrates the potential of BCHeps as sp(3)-rich motifs in drug design and provides a new surrogate for meta-substituted benzene rings in drug discovery programs.
