Imaging side effects and complications of chemotherapy and radiation therapy: a pictorial review from head to toe

Radiologists must be aware of therapeutic options, side effects, and complications to distinguish expected changes from tumor relapse.

Rapid Evaluation of Novel Therapeutic Strategies Using a 3D Collagen-Based Tissue-Like Model

Summary: A reproducible and commercially available 3D cell collagen-based model has been developed for studying the influence of the matrix on nanoagent uptake and radiation effects, offering tunable oxygen concentration and homogeneous cell density. The model allows investigation of hypoxic effects and visualization of living cells in real-time using second harmonic imaging microscopy. The disaggregation of the collagen matrix enables recovery of cells without damage, facilitating single cell analysis and quantification of treatment effects.

Theranostic AGuIX nanoparticles as radiosensitizer: A phase I, dose-escalation study in patients with multiple brain metastases (NANO-RAD trial)

Summary: The study assessed the safety and feasibility of combining AGuIX nanoparticles with whole brain radiotherapy for patients with brain metastases, showing no dose-limiting toxic effects and clinical benefits for most patients. Efficient targeting of metastases and correlation between contrast enhancement and tumor response were observed, supporting a potential radiosensitizing effect. Ongoing phase II studies will further evaluate efficacy.

Quantifying nanotherapeutic penetration using a hydrogel-based microsystem as a new 3D in vitro platform

Summary: This study presents a simple yet highly flexible 3D model microsystem consisting of agarose-based microwells for formation of reproducible spheroids, providing a promising in vitro platform for investigation of nanotherapeutic transport. The system allows spatial and temporal resolution of the penetration and distribution of nanoparticles within multicellular tumor spheroids, demonstrating differences in intra and extracellular localization of nanoparticles in 2D and 3D settings.
