A ratiometric fluorescent biosensing platform for ultrasensitive detection of Salmonella typhimurium via CRISPR/Cas12a and silver nanoclusters

Summary: We developed a versatile biosensing platform, SCENT-Cas (Silver nanoCluster Empowered Nucleic acids Test using CRISPR/Cas12a), for ultrasensitive detection of pathogenic bacteria. This platform used the isothermal amplification of the invA gene of Salmonella typhimurium to trigger trans-cleavage of CRISPR/Cas12a. The trans-cleavage was then utilized to degrade single-stranded DNA and create a ratiometric fluorescence biosensing platform. This method enabled the detection of 1 CFU/mL S. typhi with a dynamic range from 1 to 108 CFU/mL, and showed promise for real food sample testing.

Detection of cancer-associated miRNA using a fluorescence switch of AgNC@NA and guanine-rich overhang sequences

Summary: DNA-templated silver nanoclusters (AgNC@DNA) are a new type of nanomaterial with advantageous optical properties. The fluorescence of nanoclusters can be tuned using DNA overhangs. In this study, we explored the properties of AgNCs manufactured on a short single-stranded (dC)(12) with adjacent G-rich sequences (dG(N)) added. Adding a G-rich overhang resulted in a significant change in fluorescence, with the emergence of two new emissive states. This property was utilized to design biosensors capable of detecting specific nucleic acid sequences, and the detection switch showed sensitivity and specificity for a prostate cancer-related miRNA sequence.

Traditional wound dressings are not sufficient for the whole wound healing process, so there is a need to develop new wound dressings with multiple functions. Nano-silver and naturally derived polysaccharides have shown great potential in wound healing due to their antibacterial properties, good biocompatibility, and affordability.

In this study, biocompatible DNA was used as a building material to fabricate antimicrobial materials, which effectively inhibit bacterial growth without affecting cell proliferation. Particularly, Y-Ag spray can expedite the process of wound healing.

Label-free and dual-mode biosensor for HPV DNA based on DNA/silver nanoclusters and G-quadruplex/hemin DNAzyme

Summary: A label-free and fluorimetric/colorimetric dual-mode sensing strategy was developed for the quantitative determination of HPV DNA. By integrating fluorescent DNA-silver nanoclusters and G-quadruplex/hemin DNAzyme, this strategy improved the diversity and accuracy of analysis. It has been successfully applied to HPV DNA analysis in different real samples with excellent results, providing a universal strategy for various kinds of virus analysis.

Influence of silver ion release on the inactivation of antibiotic resistant bacteria using light-activated silver nanoparticles

Summary: This study confirms the synergistic effect between silver nanoparticles (AgNPs) and photosensitizers (PSs) in the light-mediated oxidation process, and shows that polyethyleneimine (PEI) coated AgNPs can enhance antibacterial activity. The most effective material was found to be PpIX-AgNPs, achieving better inactivation of multidrug resistant bacteria.

Summary: The fluorescence emission of DNA-scaffolded silver nanoclusters (AgNCs) can be modulated through confined strand displacement and transient concatenate ligation for amplifiable biosensing of a DNA segment related to SARS-CoV-2. A three-way DNA device (3WDD) is designed with three stem-loop structural hairpins and two linkers, where an orange-emitting AgNC (oAgNC) is stably clustered. The presence of the DNA segment initiates toehold-mediated strand displacement and repeatable recycling amplification, resulting in the opening of the oAgNC-hosted hairpin loops and significant fluorescence decay.
