Diselenide-Based Dual-Responsive Prodrug as Pyroptosis Inducer Potentiates Cancer Immunotherapy

Shu-Cheng Wan et al.

Summary: Pyroptosis triggers antitumor immunity and this study introduces a dual-responsive diselenide-based pyroptosis inducer that has excellent tumor microenvironment targeting effects. It can remodel the immunostimulated tumor microenvironment and trigger a robust immune response. Combined with αPD-1 therapy, it effectively inhibits the growth of remote tumors and prolongs survival.

Nano-ultrasonic Contrast Agent for Chemoimmunotherapy of Breast Cancer by Immune Metabolism Reprogramming and Tumor Autophagy

Xupeng Yang et al.

Summary: This study determines that reprogramming the metabolism of immature dendritic cells (iDCs) and tumor-associated macrophages (TAMs) can enhance the ratio of mature dendritic cells (mDCs) and proinflammatory macrophages, thus improving the immune response against triple-negative breast cancer. The findings provide a candidate method for monitoring treatment and augmenting antitumor chemoimmunotherapy by suppressing tumor cell autophagy and reprogramming immunocyte metabolism.

Immunogenic cell stress and death

Guido Kroemer et al.

Summary: Immunogenic cell death (ICD) plays an important role in therapy and disease, as dying mammalian cells release signals that interact with the host to determine the immunological correlates of cellular stress and death. ICD is crucial in immunosurveillance and is related to strategies evolved by pathogens and cancer cells. Additionally, normal cell death can also initiate antigen-specific immune responses.

Yanhua Zhai et al.

Summary: In this study, self-activated arsenic manganite nanohybrids were developed for high-contrast magnetic resonance imaging (MRI) and arsenotherapy in triple-negative breast cancer (TNBC). The nanohybrids enabled precise tumor targeting, delivered high levels of arsenic trioxide (ATO) to the tumor cells, and exhibited potent antitumor efficacy. The nanohybrids also showed synergistic effects when combined with thermotherapy and induced immunogenic cell death (ICD) to activate the tumor microenvironment and enhance the antitumor immune response.

Photo-Reduction with NIR Light of Nucleus-Targetingt Pt-IV Nanoparticles for Combined Tumor-Targeted Chemotherapy and Photodynamic Immunotherapy

Dengshuai Wei et al.

Summary: The development of Pt-IV prodrugs selectively reduced into their Pt-II active species within cancerous cells has gained increasing attention. Despite progress, compounds are mostly excited using UV or blue light, limiting treatment for deep or large tumors. Pt-IV-functionalized nanoparticles excited at 808 nm within the MR region show potential for multi-modal treatment in eradicating tumors.
