Design of Light-Activated Nanoplatform through Boosting Eat Me Signals for Improved CD47-Blocking Immunotherapy

Yan Lu et al.

Summary: The authors propose a light-activated nanoplatform that enhances immunogenic cell death and improves CD47-blocking immunotherapy. This nanoplatform can release eat me signals and perform phototherapy when exposed to near-infrared laser irradiation. In vitro and in vivo experiments demonstrate that this strategy effectively induces synergistic antitumor immune responses.

Trinity immune enhancing nanoparticles for boosting antitumor immune responses of immunogenic chemotherapy

Yudi Xu et al.

Summary: A novel trinity immune enhancing nanoparticle (TIENs) was proposed to boost antitumor immune responses of chemo agents, showing promising results in promoting antigen capturing, activating APCs, and antigen cross-presentation. In murine tumor models, TIENs greatly enhanced the antitumor effects and immune responses of oxaliplatin and doxorubicin.

Tumor microenvironment-activated therapeutic peptide-conjugated prodrug nanoparticles for enhanced tumor penetration and local T cell activation in the tumor microenvironment

Xianghui Zhu et al.

Summary: Nanomedicine-based chemoimmunotherapy has potential for enhanced tumor penetration and immunotherapeutic effects. A TME-activable prodrug nanoparticle was developed to co-deliver DOX and (D)PPA antagonist, inhibiting regulatory T cells and promoting immune response to prevent tumor recurrence and metastasis.

Radiotherapy, immunotherapy, and the tumour microenvironment: Turning an immunosuppressive milieu into a therapeutic opportunity

N. E. Donlon et al.

Summary: Immune checkpoint blockade has significantly changed the treatment of solid tumors, but many patients do not respond. Combining radiation therapy with immunotherapy may have synergistic effects by remodeling the tumor microenvironment, but careful control of dose and fractionation is essential for safety and efficacy. Ongoing efforts to combine radiation, immunotherapy, and DNA damage response inhibitors aim to achieve a beneficial balance between the immunogenic and tolerogenic effects of radiation on the immune microenvironment.

Enhanced tumour penetration and prolonged circulation in blood of polyzwitterion-drug conjugates with cell-membrane affinity

Siqin Chen et al.

Summary: Conjugates of small-molecule anticancer drugs with a polyzwitterion that has negligible interaction with proteins and weak interaction with phospholipids can effectively eradicate large tumours and patient-derived tumour xenografts in mice by facilitating their transcytosis-mediated extravasation and infiltration into tumours, showcasing potential for translational anticancer nanomedicines.

Design and Synthesis of New PEGylated Polydopamine-Based Nanoconstructs Bearing ROS-Responsive Linkers and a Photosensitizer for Bimodal Photothermal and Photodynamic Therapies against Cancer

Islam Zmerli et al.

Summary: Polydopamine nanoparticles have been considered for their multifunctional properties including photothermal and photodynamic activities. In this study, PDA NPs were modified with SH-PEG polymers bearing a reactive oxygen species-cleavable linker coupled to trisulfonatedtetraphenylporphyrin for PDT applications. The efficient grafting of PS-conjugated PEG on PDA was demonstrated in situ and the photoinduced release of PS was confirmed by H-1 NMR. In vitro experiments on human esophageal cells showed the strong synergistic effect of combining PTT and PDT within this nanoplatform.
