Structural insights and shedding light on preferential interactions of dietary flavonoids with G-quadruplex DNA structures: A new horizon

Summary: G-quadruplex is a unique structure in nucleic acids that exists widely in human genome and has attracted great attention in therapeutic investigations. Targeting G-quadruplex structure is a new strategy for drug development. Flavonoids, which are present in various dietary plant-based products, have shown high pharmacological effectiveness with minimal cytotoxicity, making them potential alternatives to synthetic therapeutic medicines. Research on the interaction between G-quadruplex and flavonoids is expected to provide newer perspectives for the development of novel therapeutic agents.

Summary: Near linear-scaling density functional theory (LS-DFT) methods were used to study the interaction between two isomers of the [Mo(eta(3)-C3H5)Br(CO)(2)(phen)] metal complex and DNA G-quadruplexes (GQ). The results showed that the axial isomer is more stable in the presence of solvent, and it interacts with GQ through end-stacking. On the other hand, the equatorial isomer prefers to interact with GQ through intercalation, with the ancillary ligands also playing a role in the interaction.

Summary: The GGGGCC (G(4)C(2)) repeat expansion in the C9orf72 gene is known to cause neurodegenerative diseases, and dipeptide repeat proteins produced through RAN translation are considered potential drivers for these diseases. The compound TMPyP4 has been shown to inhibit the translation of G(4)C(2) repeats in C9orf72, likely through stalling elongating ribosomes on the repeat RNA. This reveals a novel mode of action for TMPyP4 as an inhibitor of G(4)C(2) repeat translation elongation.
