The most promising method for its production is through microbial fermentation, particularly using yeasts, which have the ability to produce 2-PE using l-Phe as precursor. Some agro-industrial waste and by-products with high nutritional value can be utilized as suitable media for microbial growth, including the production of 2-PE through yeast fermentation.

Investigation of potential inhibitor properties of violacein against HIV-1 RT and CoV-2 Spike RBD:ACE-2

Summary: A bacterium producing violacein was isolated from a mud sample near a hot spring on Kumbet Plateau. The crude violacein pigments were separated and purified, and their inhibitory effects on SARS-CoV-2 and HIV-1 were investigated. The study found that violacein and deoxyviolacein effectively bind to key receptors and inhibit the viruses.

Valorization of Low-Cost Substrates for the Production of Odd Chain Fatty Acids by the Oleaginous Yeast Yarrowia lipolytica

Summary: Odd-chain fatty acids (OCFAs) have diverse applications in various industries, and Yarrowia lipolytica is a promising microorganism for their production. This study used sugar beet molasses and crude glycerol as carbon sources to reduce the cost of oil production by Y. lipolytica, and investigated the effects of medium composition on biomass production, lipid content, and OCFAs profile. The results showed that the fermentation process could be scaled up and fed-batch co-feeding increased OCFA accumulation in Y. lipolytica by 56% compared to batch cultures.

A Golden-Gate Based Cloning Toolkit to Build Violacein Pathway Libraries in Yarrowia lipolytica

Summary: Violacein, a naturally occurring anticancer therapeutic compound, was produced in Y. lipolytica using a library of strains constructed with Golden Gate assembly method. Gene expression analysis revealed that strong expression of VioB, VioC, and VioD favored violacein production, while weak expression of VioD was associated with high deoxyviolacein production. By optimizing culture conditions, violacein reached a maximum of 70.04 mg/L with 5.28 mg/L of deoxyviolacein in shake flasks.

Biosensor-Assisted Adaptive Laboratory Evolution for Violacein Production

Summary: The study utilized adaptive laboratory evolution (ALE) strategy and biosensor-assisted selection to enhance violacein production, resulting in a population capable of efficiently synthesizing tryptophan and identified beneficial point mutations for overproduction through whole-genome sequencing. The biosensor-assisted ALE strategy was shown to be effective in rapidly and selectively evolving producers for high violacein production.
