Enhanced Antifungal Activity of Amphotericin B Bound to Albumin: A ?Trojan Horse? Effect of the Protein

Ewa Grela et al.

Summary: We report a new method using albumin (BSA) to formulate Amphotericin B (AmB), which is highly effective against Candida albicans at low concentrations, reducing toxicity to patients. Molecular spectroscopy and imaging techniques reveal that the AmB-BSA complex readily binds to fungal cell membranes and does not form potentially toxic drug aggregates.

Amphotericin B assembles into seven-molecule ion channels: An NMR and molecular dynamics study

Yuichi Umegawa et al.

Summary: In this study, the three-dimensional structure of the molecular assemblies formed by Amphotericin B in membranes was experimentally elucidated using spectroscopy and simulations. The observed structure explains the drug's structure-activity relationships and will contribute to improving drug efficacy and reducing adverse effects.

Sixty years of Amphotericin B: An Overview of the Main Antifungal Agent Used to Treat Invasive Fungal Infections

Francelise B. Cavassin et al.

Summary: Polyenes, the oldest family of antifungal drugs introduced in the late 1950s, remain useful in the treatment of invasive fungal diseases due to their broad spectrum of activity, low resistance rate, and excellent clinical and pharmacological action despite their toxic potential. Focus has been placed on developing new products to minimize the known toxicity of the conventional drug.

Self-assembly, stability and conductance of amphotericin B channels: bridging the gap between structure and function

Joanna Zielinska et al.

Summary: This study investigates the structural and thermodynamic aspects of Amphotericin B channel formation, confirming the formation of stable octameric double-length channels and explaining the effects of sterols on channel stability and observed cation selectivity. Through fluorescence lifetime imaging and molecular simulations, the research traces the process of channel assembly and ion permeation, shedding light on the role of tail hydroxyl in the assembly of functional channels and possible origins of cholesterol-ergosterol selectivity.
