Summary: Compound 1 is a novel brain-type glycogen phosphorylase inhibitor with protective effects on ischemic brain injury. It alleviates hypoxic-ischemic injury in astrocytes by improving cell viability and reducing LDH leakage, intracellular glucose content, and post-ischemic ROS level. It also improves cellular energy metabolism, reduces anaerobic glycolysis, and inhibits apoptosis.

Discovery of novel heterocyclic derivatives as potential glycogen phosphorylase inhibitors with a cardioprotective effect

Summary: This study evaluates the effect of GP inhibitor as a potential pharmaceutical target on MI/R injury. The researchers designed and synthesized four different structural types of novel compounds, obtaining 31 novel GP inhibitors. Compound IIIh showed strong inhibitory activity against multiple subtypes of GP and exhibited obvious cardioprotective effect on MI/R injury mice in vivo. The discovery of compound IIIh provides a new strategy for developing novel GP inhibitors with myocardial ischemia protection.
